הפגיעה ACL היא בעיה בדרך נתקלה באנשים פעילים. אפילו דמעות חלקיות של רצועה בברך להוביל תוך מפרק זה לליקויים ביומכניים הפוגעים בתפקוד ויציבות. אפשרויות הנוכחיות לטיפול בדמעות ACL החלקיים נעות בין ניהול שמרני, שמרני לאפשרויות כירורגיות מרובות, כגון: שינוי תרמי, תיקון יחיד צרור, שחזור מלא, ובנייה מחדש של החלק הפגום של רצועת הילידים. מחקרים מעטים, אם בכלל, הוכיחו בכל שיטה יחידה עבור ניהול להיות מעולה באופן עקבי, ובמקרים רבים חולים ממשיכים להפגין חוסר יציבות מתמשכת ומחלות רקע אחרות. מטרתו של מחקר זה היא לזהות מקור תא פוטנציאל לניצול בפיתוח של תיקון רקמות מהונדסות שיכול להיות מיושם בתיקון של ACL קרוע באופן חלקי. פרוטוקול רומן פותח להרחבת התאים שמקורם patieלילות עוברים שחזור ACL. כדי לבודד את התאים, רקמות hACL טחונות שהושגו במהלך שחזור ACL היו מתעכל בפתרון Collagenase. הרחבה בוצעה באמצעות מדיום DMEM/F12 תוספת 10% בסרום שור העובר (FBS) ו -1% פניצילין / סטרפטומיצין (P / S). התאים אוחסנו לאחר מכן ב-80 º C או בחנקן נוזלי בהקפאה בינונית בהיקף של DMSO, FBS ובינוני ההתרחבות. לאחר הפשרה, תאי hACL נגזרים היו אז זורעים על פיגום רקמה מהונדסת, PLAGA (לקטית, חומצה גליקולית שיתוף פולי) וקלקר תרבות רקמות שליטה (TCPS). לאחר 7 ימים, SEM בוצע כדי להשוות הידבקות סלולרית לPLAGA לעומת TCPS השליטה. מורפולוגיה תאית הוערכה באמצעות מכתים immunofluorescence.
