CD4 + Effektor-T-Zellen reagieren auf fremde Antigen durch Sekretion von Zytokinen, die viele Aspekte der Immunität regulieren und kann auch direkt helfen, B-Zellen, Antikörper zu produzieren. CD8 + zytotoxischer T-Zellen (CTLs) können auch Fremdantigen reagieren durch Sekretion von Zytokinen und spielt eine zentrale Rolle bei der Abtötung von Zellen direkt ein fremdes Antigen exprimiert. Die grundlegende Interaktion, die diese T-Zell-Effektorfunktionen initiiert beinhaltet die Interaktion des T-Zell-Rezeptor (TCR) mit fremden Peptiden auf MHC-Molekülen auf der Oberfläche von Zellen angezeigt. CD4 + T-Zellen erkennen Peptide an MHC-Klasse-II-Molekülen auf Antigen-präsentierenden Zellen und CD8 + T-Zellen erkennen das Peptiden auf MHC-Klasse-I-Moleküle, die typischerweise auf Mikrobe infizierten Zellen angezeigt werden, angezeigt. Die FTA-Test ist daher ein beispielloses Niveau von T-Zell-Antwort-Effektor Beurteilung in vivo. Hier beschreiben wir im Detail die Konstruktion einer FTA und zeigen, wie sie zur Bestimmung der T-Zellantworten in vivo angewendet werden. Das Verfahren beschreibt die Konstruktion eines FTA 252 erkennbaren Zellcluster durch die Verwendung von drei Vitalfarbstoffen, 6 Wiederholungen der 42 Zellcluster mit MHC-Klasse-I und-II-bindenden Peptiden gepulst aufweist, aufweisend. Die Markierung von Zellclustern 42 erfolgt in 10 ml-Röhrchen mit konischem Boden und es ist hilfreich, um diese in einem Reagenzglasgestell lag wie in Tabelle 1 dargestellt. Dieses Verfahren kann für eine kleinere Anzahl von erkennbaren Cluster eingestellt, wie durch die Verringerung der Menge der Markierung erforderlich jeder Farbstoff 1 durchgeführt. Wir zeigen den Nutzen des Tests zeigen, wie es Respons messenes durch rekombinante Pocken-Virus-Impfung gegen mehrere Epitope in einer kleinen Kohorte von Mäusen erzeugt. Hier beschreiben wir die Verwendung von fluoreszierenden Zielarrays (FTA)-Technologie, die Vitalfarbstoffe wie Carboxyfluoreszein Succinimidylester (CFSE), violetten Laser erregbar Farbstoffe (CellTrace Violett: CTV) nutzt und rote Laser erregbar Farbstoffe (Dye Cell Proliferation eFluor 670: CPD ) Etiketten Maus Lymphozyten in&gt; 250 erkennbar fluoreszierenden Zellhaufen kombinato. Zellhaufen innerhalb dieser Freihandelsabkommen mit Haupthistokompatibilitäts (MHC)-Klasse-I und MHC-Klasse-II-bindenden Peptiden gepulst werden und dienen als Zielzellen für CD8 + und CD4 + T-Zellen, die jeweils damit. Diese FTA Zellen lebensfähig bleiben und voll funktionsfähig und kann daher in Mäuse verabreicht werden, um Beurteilung der CD8 + T-Zell-vermittelte Abtötung von Zielzellen und FTA CD4 + T-Zell-meditierten hel ermöglichenp von freien Zielzellen B-Zellen in Echtzeit in vivo durch Durchflusszytometrie. Da&gt; 250 Zielzellen auf einmal geprüft werden, ermöglicht die Technik der Überwachung der T-Zell-Antworten gegen verschiedene Antigen-Epitope in verschiedenen Konzentrationen und in mehreren Wiederholungen. Als solche kann die Technik T-Zell-Antworten sowohl auf quantitative (zB der kumulativen Größe der Antwort) und eine qualitative (z. B.. Funktionelle Avidität und Epitop-Kreuzreaktivität der Antwort-) Pegel zu messen. Hier beschreiben wir, wie diese Freihandelsabkommen sind konstruiert und geben ein Beispiel, wie sie angewendet werden, um T-Zell-Antworten durch einen rekombinanten Pockenvirus-Impfstoff induziert beurteilen.
