Het muismodel van TEVG is een waardevol hulpmiddel om cellulaire en moleculaire mechanismen van neotissue vorming en de ontwikkeling van stenose bestuderen. Aangeboren hartafwijkingen zijn ernstige aandoeningen die bijna 8% van de levendgeborenen in de Verenigde Staten beïnvloeden. Ongeveer 25% van de zuigelingen met aangeboren hartafwijkingen of 2,4 per 1000 levend geboren, vereisen invasieve behandeling in het eerste jaar van hun leven 1. De meest effectieve behandeling voor aangeboren hartafwijkingen is reconstructieve chirurgie. Helaas, complicaties als gevolg van het gebruik van thans beschikbare vasculaire leidingen zijn de belangrijkste oorzaak van postoperatieve morbiditeit en mortaliteit. Om dit probleem aan te pakken, ontwikkelden we de eerste tissue engineered vaattransplantaten (TEVGs) voor klinisch gebruik 2. TEVGs werden opgebouwd uit biologisch afbreekbare polyester buizen bezaaid met autoloog beenmerg-cellen (BM-MNO&#39;s) en geïmplanteerd als veneuze leidingen voor congenitale hartchirurgie. De resultaten toonden een uitstekende doorgankelijkheid bij 1-3 jaar follow-up, maar met significante incidentie van stenose <sinschrijven&gt; 3,4. Het was duidelijk dat een beter begrip van vasculaire neotissue vorming en het mechanisme achter de ontwikkeling van TEVG stenose nodig is. Om beter inzicht in de ontwikkeling van TEVGs en het mechanisme van stenose ontwikkeling, werd een schaap model gecreëerd 5,6. In dit model, de TEVGs succes getransformeerd in levende schepen en waren vergelijkbaar in zowel morfologie en functie van natief aderen. Dit gebruik van een groot diermodel was een goede eerste stap deze belangrijke pre-klinische informatie die klinische gebruik van TEVGs geholpen. Echter volledig begrip van de cellulaire en moleculaire mechanismen van vasculaire neotissue formatie TEVGs met grote diermodellen beperkt gevolg van beperkingen in de moleculaire karakterisatie van vasculaire cel fenotypes gebrek aan soortspecifieke moleculaire technieken. Om deze tekortkomingen te overwinnen, een muizenmodel van TEVGs werd ontwikkeld op grond van de snelle vooruitgang in de muis genetica en hun uitgebreide molecular karakterisering met het extra voordeel van een kortere tijdschaal. Biologisch afbreekbare scaffolds geënt beenmerg mononucleaire cellen (BMCs) worden vaak gebruikt voor reconstructieve chirurgie aangeboren hartafwijkingen te behandelen. De lange-termijn klinische resultaten toonden uitstekende doorgankelijkheid, echter met significante incidentie van stenose.
