Die Daten zeigten einen Anstieg in der Dicke der Wand in den LV myocarditic Tieren, entsprechend einer Abnahme der LV enddiastolischen Volumens, des Schlagvolumens und der Auswurffraktion, verglichen mit diesen funktionellen Parameter in gesunden Mäusen.

noninvasive assessment of cardiac abnormalities in experimental autoimmune myocarditis by magnetic resonance microscopy imaging in the mouse

Myokarditis ist eine Entzündung des Herzmuskels, aber nur ca. 10% der Betroffenen zeigen klinische Manifestationen der Erkrankung. Um die Immun Ereignisse der myokardialen Verletzung zu untersuchen, wurden verschiedene Mausmodelle der Myokarditis weit verbreitet. Diese Studie umfasste experimentellen autoimmunen Myokarditis (EAM) mit Herz Myosin schwere Kette (Myhc)-α 334-352 in A / J-Mäusen; die betroffenen Tiere entwickeln lymphatischer Myokarditis, aber ohne offensichtliche klinische Zeichen. In diesem Modell wird der Nutzen der Magnetresonanzmikroskopie (MRM) als eine nicht-invasive Modalität der Herz strukturelle und funktionelle Veränderungen bei Tieren mit Myhc-α 334-352 immunisiert bestimmen dargestellt. EAM und gesunden Mäusen wurden mit einem 9.4 T (400 MHz) 89 mm vertikale Kernbohrung Scanner mit einem 4 cm Tausendfüßler Hochfrequenz-Bildgebung Sonde und 100 G / cm Dreifachachse Gradienten ausgestattet abgebildet. Cardiac Bilder wurden von betäubten Tieren erworben mit einem Gradienten-Echo-basierte cine Impulsfolge, und der Animals wurden von Atmung und Pulsoxymetrie überwacht. Die Analyse ergab eine Erhöhung der Dicke der Kammerwand in EAM Mäusen, mit einer entsprechenden Abnahme des Innendurchmessers der Kammern, wenn sie mit gesunden Mäusen verglichen. Die Daten legen nahe, dass morphologische und funktionelle Änderungen in den entzündeten Herzen können nicht-invasiv überwacht MRM in lebenden Tieren sein. Abschließend bietet einen Vorteil MRM Beurteilung der Progression und Regression der myokardialen Verletzung in Krankheiten, die durch infektiöse Agenzien verursacht werden, sowie als Reaktion auf Therapien.

In diesem Bericht wird der Nutzen der MRM-Technik als eine nicht-invasive Modalität, die strukturelle und funktionelle Veränderungen in den Herzen der Tiere mit EAM beeinflusst zu werden gezeigt. Myokarditis in A / J-Mäusen durch Immunisierung der Tiere mit Myhc-α von 334 bis 352 in 7 CFA induziert und die Tiere wurden am Tag 21 nach der Immunisierung an MRM Experimenten unterworfen. This study demonstrates the successful establishment of magnetic resonance microscopy imaging as a non-invasive tool to assess the cardiac abnormalities in mice affected with autoimmune myocarditis.
