Afin d&#39;étudier in vitro les interactions hôte-pathogène dans lors de l&#39;infection par E. épithéliale histolytica, il est essentiel de travailler avec des cultures bien établies des deux cellules épithéliales et trophozoïtes. Par exemple, autrefois, E. cultures histolytica habituellement avaient été mis en place en association avec certaines espèces de bactéries ou trypanosomatidés 22,23. Entamoeba histolytica est un protozoaire responsable de l&#39;amibiase humaine, une infection intestinale cellule causant l&#39;inflammation et la diarrhée. E. histolytica infecte jusqu&#39;à 50 millions de personnes chaque année, mais seulement 10% des personnes infectées développent les symptômes associés à l&#39;amibiase 1. Entamoeba histolytica est l&#39;agent causal de l&#39;amibiase humaine, une cause majeure de diarrhée et abcès du foie dans les pays tropicaux. L&#39;infection est initiée par l&#39;interaction de l&#39;agent pathogène aux cellules épithéliales de l&#39;intestin. Cette interaction conduit à la rupture des structures intercellulaires tels que les jonctions serrées (TJ). TJ assurer l&#39;étanchéité de la couche epitheliale pour séparer le tissu de l&#39;hôte à partir de lumière de l&#39;intestin. Des études récentes fournissent la preuve que la perturbation de la EhCPADH112 TJ en protéine de parasite est une condition préalable à E. invasion de histolytica qui est accompagné par un dysfonctionnement de la barrière épithéliale. Ainsi, l&#39;analyse des mécanismes moléculaires impliqués dans TJ démontage pendant E. invasion de histolytica est d&#39;une importance capitale pour améliorer notre compréhension de la pathogenèse amibiase. Cet article présente un modèle simple qui permet l&#39;évaluation des interactions initiales hôte-pathogène et le potentiel d&#39;invasion de parasites. Paramètres à analyser sont trésistance électrique ransepithelial, l&#39;interaction de EhCPADH112 avec les récepteurs de surface épithéliales, changements dans l&#39;expression et la localisation de marqueurs de jonction épithéliales et la localisation des molécules du parasite dans les cellules épithéliales.

Pour un succès E. culture histolytica, deux conditions importantes doivent être remplies: la croissance dans des conditions axéniques et récolte en phase logarithmique. Auparavant, les cultures de E. histolytica ont facilement créé en association avec certaines espèces de bactéries ou trypanosomatidés 22.
