Entamoeba histolytica ist der Erreger der menschlichen Amöbenruhr, eine Hauptursache für Durchfall und Leber Abszess in tropischen Ländern. Infektion durch Wechselwirkung des Pathogens mit Darmepithelzellen eingeleitet. Diese Wechselwirkung führt zu einer Störung der interzellulären Strukturen wie tight junctions (TJ). TJ sicherzustellen Abdichtung der Epithelschicht auf das Wirtsgewebe von Darmlumen trennen. Neuere Studien belegen, dass eine Unterbrechung der TJ durch die parasitäre Protein EhCPADH112 ist eine Voraussetzung für E. histolytica Invasion, die von epithelialen Barrierestörung begleitet wird. Eine Analyse der molekularen Mechanismen in TJ Demontage während E. beteiligten histolytica Invasion ist von größter Bedeutung für unser Verständnis der Pathogenese Amöbenruhr zu verbessern. Dieser Artikel stellt eine einfache Modell, das die Beurteilung der ersten Wirt-Pathogen-Interaktionen und den Parasiten Invasionspotential ermöglicht. Parameter analysiert werden umfassen transepithelial elektrischen Widerstand, Interaktion von EhCPADH112 mit epithelialen Oberflächenrezeptoren, Veränderungen in der Expression und Lokalisation von epithelialen Marker junctional und Lokalisierung von Parasiten Moleküle innerhalb Epithelzellen.

Für eine erfolgreiche E. histolytica Kultur, müssen zwei wichtige Bedingungen erfüllt sein: Wachstum in axenischen Bedingungen und Ernte im logarithmischen Phase. Zuvor Kulturen von E. histolytica wurden leicht in Verbindung mit bestimmten Arten von Bakterien oder Trypanosomatiden 22 etabliert. Allerdings ist es heutzutage üblich, axenischen Anbau dieser Parasiten bedeutet eine unbestimmte Subkultivierung von Amöben in einer Umgebung frei von metabolisierenden Bakterien, Pilze, Protozoe...
