För att studera in vitro värd-patogen interaktioner under epitel infektion av E. histolytica, är det viktigt att arbeta med väletablerade kulturer av både epitelceller och trophozoites. Till exempel, tidigare, E. histolytica kulturer hade vanligtvis upprättats i samband med vissa arter av bakterier eller trypanosomatids 22,23. Entamoeba histolytica är en enda cell protozo ansvarig för mänsklig amoebiasis, en tarminfektion som orsakar inflammation och diarré. E. histolytica infekterar upp till 50 miljoner människor varje år, men endast omkring 10% av smittade människor utvecklar symptom i samband med amoebiasis 1. Infektion sker vid intag av förorenad mat eller vatten som innehåller E. histolytica cystor. I tarmen, cystor producerar levande trofozoiter som följer kolon mucin och föröka 2. Trofozoiter bildar vanligtvis cystor som utsöndras via avföringen. I andra fall, och för ännu okänd anledning, trofozoiter bryter tarmens epitel lagret och invadera underliggande vävnader. I värsta fall, de går in i blodomloppet och påverkar andra organ såsom lever 3. Breaking epitelbarriären kräver avbrott i epithelial transmembran strukturer som upprätthåller celler förenade. Epithelial cellkontakter är bildade genom den apikala Junktional komplex bestående av tät (TJ) och adherensgränssnitten korsningar (AJ), och desmosomer 4. De är intracellulärt bundna av ställnings molekyler av zonula occludens (ZO) familj som ansluter adhesionsreceptorer till aktin cytoskelettet att ge ytterligare mekanisk hållfasthet till epitelet. TJ är ansvariga för att täta tarmvävnad från tarmlumen, förhindra alltför vatten och lösta ämnen läckage. Således, efter TJ störs av parasiten, är vävnaderna invaderade. E. histolytica utsöndrar flera molekyler såsom: (i) de som är involverade i vidhäftning av amöbor till Target celler 5; (II), membranaktiva faktorer som deltar i avlivning av värdceller genom exocytos, exempelvis jon-kanalbildande peptider benämnda amoebapores 6,7; och (iii) proteinaser som bryter ned extracellulära matrisproteiner, och medierar vävnadssönderfalls 5,8,9. Den cysteinproteas EhCP112 och adhesionsmolekylen EhADH112 som tillsammans bildar EhCPADH112 komplexet är två E. histolytica virulens proteiner som spelar en viktig roll vid demonteringen av TJ 10. Live trofozoiter, deras totala lysat och utsöndrade produkter inducerar molekylära förändringar i TJ komplex och funktionell störning av epiteliala barriären. I denna studie visas det att EhCP112 och EhADH112 samverka med occludin och claudin-1-proteiner som leder till internalisering och nedbrytning av cellproteiner, vilket underlättar E. histolytica ingång genom paracellulära vägen. Våra data och de som of andra grupper 11-17 tyder starkt nödvändigheten av specifika värd-patogen interaktioner som tillåter parasit invasion. Reda ut den molekylära grunden för dessa interaktioner är av största betydelse för en bättre förståelse av amoebiasis patogenes. Selektiv störning av TJ av trofozoiter, kännetecknade av ökad paracellulär permeabilitet, kan mätas genom en minskning i transepitelial elektrisk resistans (TER). Den överföring av parasitproteiner mot värd epitel kan bestämmas genom immunofluorescens färgning och konfokal laser mikroskopi, en metod som också kan avslöja co-lokalisering av amöba virulensfaktorer med epithelial junctionala markörer som anger eventuella direkta interaktioner. Entamoeba histolytica är vilka smittämnen av humant amoebiasis, en viktig orsak till diarré och lever abscess i tropiska länder. Infektion initieras genom interaktion av patogenen med tarmepitelceller. Denna interaktion leder till störningar av intercellulära strukturer som tight junctions (TJ). TJ säkra tätning av epitelskiktet att skilja värdvävnad från tarmlumen. Nya studier visar att störning av TJ från parasitprotein EhCPADH112 är en förutsättning för E. histolytica invasion som åtföljs av epitelbarriär dysfunktion. Således, analys av molekylära mekanismer som är involverade i TJ demontering under E. histolytica invasion är av största vikt för att förbättra vår förståelse av amoebiasis patogenes. I denna artikel presenteras en enkel modell som gör det möjligt att bedöma inledande värd-patogen interaktioner och parasiten invasionen potential. Parametrar som skall analyseras innefattar transepithelial elektriskt motstånd, interaktion av EhCPADH112 med epitel ytreceptorer, förändringar i uttryck och lokalisering av epiteliala junctionala markörer och lokalisering av parasit molekyler i epitelceller.

För en lyckad E. histolytica kultur, måste två viktiga villkor uppfyllas: tillväxt i axeniska förhållanden och skörd i logaritmisk fas. Tidigare, kulturer av E. histolytica var lätt etablerade i samband med vissa arter av bakterier eller trypanosomatids 22.
