Onder hen, pulmonaalklep stenose en bicuspide aortaklep gebreken zijn een vaak voorkomende vorm 3. Hartklepvervanging operatie is een routine keuze van reconstructieve chirurgie; echter complicaties zoals stenose en verkalking van de hartklep en levenslange afhankelijkheid van anticoagulantia zijn een belangrijke bron van chronische ziekte en dood 4-7. Tissue engineered hartkleppen, vooral ontceld kleppen, beginnen aan kracht winnen in klinisch gebruik van reconstructieve chirurgie met gemengde resultaten. Echter, de cellulaire en moleculaire mechanismen van de neotissue ontwikkeling, klep verdikking en stenose ontwikkeling niet uitgebreid onderzocht. Om de bovenstaande vragen te beantwoorden, ontwikkelden we een muizen heterotopic hartklep transplantatie model. Een hartklep werd geoogst van een afsluiter donor muis en getransplanteerd naar een hart donor muis. Het hart met een nieuwe klep werd heterotoop getransplanteerd naar een ontvanger muis. De getransplanteerde hart toonde zijn eigen hartslag, onafhankelijk van de hartslag van de ontvanger. De doorbloeding werd gekwantificeerd met behulp van een hoge frequentie ultrasoon systeem met een gepulste golf Doppler. De stroming door de geïmplanteerde pulmonaalklep toonde aanvoer met minimale oprispingen en de piekstroom was dicht bij 100 mm / sec.
