C. elegans is een eenvoudige genetisch modelorganisme dat bacteriën eet als bron van voedingsstoffen. De vrijlevende bodem nematode Caenorhabditis elegans is een eenvoudige en genetisch handelbaar modelorganisme gebruikt voor vele biologische vraagstukken te bestuderen. C. elegans overwegend bestaat als zelfbemestende hermafrodieten. Mannetjes worden spontaan gegenereerd door niet-scheiding van het X-chromosoom tijdens gametogenese 1,2. In aanwezigheid van overvloedig voedsel, C. elegans voortdurend te ontwikkelen door middel van vier larvale stadia tot volwassene. Temperatuur beïnvloedt ook C. elegans ontwikkeling; snellere ontwikkeling waargenomen bij hogere temperaturen. In het laboratorium, C. elegans is gekweekt bij een standaard temperatuur van 20 ° C op agar platen met zaadjes bacterie Escherichia coli (stam OP50) als voedsel 1,2. In het laatste decennium, C. elegans heeft zich ontpopt als een ongewerveld organisme om interacties tussen gastheren en ziekteverwekkers te bestuderen, met inbegrip van de afweer tegen gram-negatieve bacterie Salmonella typhimurium. Sinds C. elegans hele genoom RNAi bibliotheken beschikbaar zijn, dit protocol maakt het mogelijk om uitgebreid te screenen op C. elegans genen die beschermen tegen Salmonella en andere intestinale pathogenen met behulp van genoom-brede RNAi bibliotheken.

Bij 20 ° C, de gemiddelde levensduur van wild-type N2 wormen is 17 dagen (Figuur 2A en tabel 2).
