Kürzlich haben wir gezeigt, bakterielle Biofilmbildung Dynamik in einem Mikrofluidik-Vorrichtung, die porösen Medien 1 nachahmt. Bakterielle Biofilme sind im Wesentlichen Kolonien von Bakterien, die Oberfläche aggregiert von extrazellulären polymeren Substanzen (EPS) 2-4 eingeschlossen sind. Diese dünnen Schichten von Bakterien bilden können, in fast jede denkbare Nische, die von glatten Oberflächen auf die viel komplexer Lebensraum von porösen Medien. Valiei et al. 1 verwendet eine Mikrofluidik-Vorrichtung mit einem Array von Mikrosäulen, um eine poröse Struktur zu simulieren und Medien untersucht Biofilmbildung bei dieser Vorrichtung als eine Funktion der Fluidströmungsrate. Sie fanden, dass in einer bestimmten Strömungsregime begann Faden Biofilme als Luftschlangen bekannt, zwischen verschiedenen Säulen entstehen. Schlangen können sich in einem angebunden werden, oder an beiden Enden an festen Oberflächen, aber der Rest der Struktur ist in Flüssigkeit suspendiert. Vor kurzem haben mehrere Forscher die Dynamik der Streamer Bildung untersucht. Yazdi et al. 5 zeigte, dass die Luftschlangen können in Wirbelströme aus einer oszillierenden Blase Ursprung zu bilden. In einem anderen Experiment Rusconi et al. 6 untersucht die Wirkung der Kanalkrümmung und Kanalgeometrie auf die Bildung von Streamern. Sie fanden heraus, dass die Luftschlangen in gekrümmten Abschnitten von Mikrokanälen zu bilden, und Streamer Morphologie an Beweglichkeit zusammen. Neuere Forschungen haben gezeigt, dass Luftschlangen können weitreichende Auswirkungen in verschiedenen natürlichen und künstlichen Szenarien haben, wie sie als Vorläufer zur Bildung von reifen Strukturen in porösen Schnittstellen fungieren können, führen zu schnellen und katastrophalen Biofilm Proliferation in einer biomedizinischen Systemen und auch zu erheblichen Fluss Struktur-Wechselwirkungen, etc 1,7-9. Luftschlangen bilden Biofilm ich oftn komplexen Lebensräumen wie poröse Medien. Verständnis Biofilmwachstum in porösen Medien-Umgebung relevant zu mehreren ökologischen und industriellen Prozessen wie der biologischen Abwasserbehandlung 10, die Aufrechterhaltung Bohrloch Integrität in Situationen, wie beispielsweise CO 2 Erfassung 11 und ein Verstopfen der Poren im Boden 12. Beobachten Biofilmbildung in solchen komplexen Lebensräumen oft schwierig sein, aufgrund der Opazität des porösen Mediums. In solchen Situationen können Mikrofluidik basierend porösen Medien Plattformen sehr vorteilhaft erweisen, da sie in Echtzeit und in situ-Überwachung zu ermöglichen. Ein weiterer Vorteil der Mikrofluidik ist die Möglichkeit, mehrere Bioreaktoren auf einem Bio-Mikrofluidik-Plattform zu bauen und gleichzeitig ermöglichen die Online-Überwachung und / oder Einbau von Sensoren. Die Flexibilität, mehrere Laborversuche in einem Gerät und die Fähigkeit, signifikante relevanten Daten für eine präzise statistische Analyse zu sammeln implementieren ist ein wichtiger advantage von mikrofluidischen Systemen 13,14. In Zusammenhang mit der obigen Diskussion würde das Verständnis Streamer Bildung Dynamik in einem porösen Medium Umgebung vorteilhaft, mehrere Anwendungen. In dieser Studie, entwickeln wir das Protokoll für die Untersuchung von Streamer-Bildung in einem Gerät, das imitiert porösen Medien. Herstellung von mikrofluidischen Plattform, sind notwendige Schritte für die Zellkultur und Experimente beschrieben. In unseren Experimenten wurde die Wildtyp-Bakterienstamm Pseudomonas fluorescens beschäftigt. P. fluorescens, natürlich im Boden gefunden, spielt eine Schlüsselrolle bei der Aufrechterhaltung der Bodenökologie 15. Der Bakterienstamm eingesetzt worden gentechnisch verändert, um das grün fluoreszierende Protein (GFP) exprimieren konstitutiv.

Verschiedene Bakterienarten besitzen die Fähigkeit, an Oberflächen anzubringen und zu kolonisieren sie in Form von dünnen Filmen als Biofilme bezeichnet. Biofilme, die in porösen Medien wachsen relevant sind mehrere Industrie-und Umweltprozesse, wie die Abwasserbehandlung und die CO 2-Sequestrierung. Wir verwendeten Pseudomonas fluorescens, ein gram-negatives Bakterium Aerobic, um die Biofilmbildung in einem mikrofluidischen Gerät, ahmt porösen Medien zu untersuchen. Mikrofluidik-Vorrichtung besteht aus einem Array von Mikrostellen, die unter Verwendung von Soft-Lithographie hergestellt wurden. Anschließend Biofilmbildung in diesen Geräten mit Fluss wurde untersucht, und wir zeigen die Bildung von Biofilmen als Fadenluftschlangen in unserem Gerät bekannt. Unter Verwendung des oben erwähnten Mikro Protokoll wurde eine PDMS basierend mikrofluidischen Vorrichtung konstruiert. 1 zeigt die rasterelektronenmikroskopische (REM) Aufnahmen der PDMS-Gerät. 1A zeigt die Eingangsabschnitt der Vorrichtung.
