Vi har beskrivit en metod för tillämpning av IF av BE celler till att belysa de fysiologiska effekterna av riktade terapier vid dessa celler. Barretts esofagus (BE) är en metaplastisk förändring i den normala skivepitel av matstrupen och en följd av kronisk exponering för maginnehåll följer av gastroesofageal refluxsjukdom (GERD) 1. BE är tänkt att vara en skyddande mekanism som svar på GERD emellertid närvaron av BE förlänar en ökad risk för esofagusadenokarcinom (EAC), en sjukdom som uppbär en signifikant dålig överlevnad 1. Aktuella uppskattningar tyder på att upp till 5,6% av den amerikanska befolkningen har BE, men som BE är ofta symptomfri, är det tänkt att majoriteten av BE förblir odiagnostiserade 2. Eftersom incidensen av både GERD och EAC har sett fortsatt tillväxt 3, har det blivit viktigt att förstå de molekylära mekanismer som är inblandade i utvecklingen av BE till EAC, särskilt eftersom denna information skulle kunna ge terapeutiska strategier för att förhindra progression av BE till EAC. 1. Kronisk inflammation, skada, och genotoxiska skador till följd av långvarig exponering för maginnehåll utövar selektivt tryck på BE lesionen främja neoplastisk progression till EAC. Ett antal genetiska förändringar har identifierats under BE till EAC progression. Trots detta finns det en tydlig brist på förståelse om de exakta förändringarna i cellsignalering och efterföljande effekter på cellstruktur och funktion. Förevigat in vitro cellinjer är användbara verktyg för att studera effekter av cellsignalering, särskilt i att undersöka effekterna av riktade terapeutiska föreningar. Den senaste tidens kommersiella tillgängligheten av odödliga BE cellinjer möjliggör sådana studier. Trots hög kapacitet analyser, såsom de allmänt använda lönsamhetsanalyser, kan vara värdefull för bedömningen av effekterna av riktade terapier vid celldelning och survival 4-6, dessa analyser inte är användbara för att förhöra effekterna av cellsignalering vid cell morfologi. Immunofluorescerande färgning (IF) är en användbar teknik för att undersöka effekterna av riktade terapier vid de egenskaper morfologiska, tillväxt och överlevnad av celler och proteiner som är involverade. Vårt laboratorium har anpassat dessa metoder mot odödliga BE cellinjer, med hjälp av IF för att utvärdera effekterna av ett kliniskt tillgängliga läkemedel på BE cellinjer i hopp om att avgränsa en eventuell chemopreventative behandling och biomarkör för läkemedelsbehandling 7. På samma sätt, tillämpning av dessa tekniker i EAC, BE och förevigade esofagus cellinjer har avgränsat kritiska rön angående BE till EAC progression 8-10. Vi finner IF analys av BE celler som behandlats med riktade terapier har värde, vilket möjliggör karakterisering av förändringar i cellstruktur och protein lokalisering. Här presenterar vi våra metoder för IF av odödliga BE celler till characterize drogbehandling.

Esofagus adenocarcinom (EAC) har en total överlevnad på mindre än 17% och förekomsten av EAC har ökat dramatiskt under de senaste två decennierna. En av de primära riskfaktorerna för EAC är Barretts esofagus (BE), en metaplastisk förändring av den normala skvamösa esophagus som svar på kronisk halsbränna. Trots den väl etablerade samband mellan EAC och BE, förhör av de molekylära händelser, särskilt förändrade signalsystem som involverar utvecklingen av BE till EAC, är dåligt kända. Mycket av detta beror på avsaknaden av lämpliga in vitro-modeller finns tillgängliga för att studera dessa sjukdomar. Nyligen har förevigat BE cellinjer blivit kommersiellt tillgängliga som medger in vitro-studier av BE. Här presenterar vi en metod för immunofluorescens färgning av odödliga BE cellinjer, vilket gör in vitro karaktärisering av cellsignalering och struktur efter exponering för terapeutiska föreningar. Tillämpning av dessa tekniker kommer att HELp utvecklar insikt i de mekanismer som är involverade i BE till EAC progression och ge potentiella vägar för behandling och förebyggande av EAC.
