Latex reagentia kunnen ook oversteken reageren als de antistoffen niet goed zijn uitgelijnd op de latex deeltjes; deze worden gedetecteerd als QC mislukkingen. In onze ervaring, strenge QC is voor het succes van deze werkwijze, ook wanneer de handel verkrijgbare antisera worden voor productie. QC duurt ongeveer 15 minuten per reagens en voert een reeks passende QC isolaten zoals hierboven beschreven. De hier beschreven methode resulteert in reagentia die een positieve reactie geven goed widun de testperiode. In onze ervaring, valse positieven zijn zeldzaam in de praktijk (gegevens niet getoond). Occasionele vals-negatieve reacties gezien worden; meestal deze betrekking hebben op bekende problemen met een bepaalde antiserum (bijvoorbeeld kan serogroep 6 isolaten niet reageren met zwembad B antiserum), of omlaag regulering van capsule expressie door het isolaat. De serotypering algrorithm door de fabrikant is belangrijk, zoals in de meeste gevallen een vals positieve of negatieve reactie zal leiden tot een &quot;blind end &#39;resultaat. In deze gevallen en wanneer reacties zijn moeilijk te interpreteren, adviseren wij herhaal-testen en / of het testen van een alternatieve methode zoals het Queliung reactie. Sommige problemen oplossen is soms nodig om bevredigende latex reagentia maken. Antilichaam beklede latex deeltjes worden vaak gebruikt in diagnostische microbiologie voor herkenning of serotypering van verschillende microben 15,20. Als zodanig kan de hier beschreven werkwijze geschikt voor het bereiden latex reagentia voor andere doeleinden, mits geschikte antisera is vrij en adequate QC procedures worden vastgesteld.

Streptococcus pneumoniae (de pneumokok) is een belangrijke oorzaak van morbiditeit en mortaliteit bij kinderen onder de leeftijd van vijf jaar de hele wereld, vooral in arme omgevingen 1,2. Pneumokokkeninfecties varieert van lokale infecties tot levensbedreigende aandoeningen zoals pneumonie, sepsis en meningitis 1,2. Meer dan 90 serotypen van pneumokokken geïdentificeerd op basis van verschillen in hun capsulair polysaccharide 3. Voor vaccins gericht de capsulaire polysaccharide en bescherming van de grote serotypen die invasieve ziekten veroorzaken 4. Serotypering pneumokokken isolaten is van belang voor de beoordeling van de impact van vaccinatie op de wagen en de ziekte alsmede het verstrekken van ruimere epidemiologische informatie 5,6. De huidige &#39;gouden standaard&#39; methode voor pneumokokken serotypering is de Queliung test, maar het is tijdrovend en vereist enige vaardigheid om perform 7. Latex agglutinatie is een alternatieve methode voor serotypering aanbevolen door de World Health Organization 7. capsular serotyping of streptococcus pneumoniae by latex agglutination

Latex agglutination reagents are widely used in microbial diagnosis, identification and serotyping. Streptococcus pneumoniae (the pneumococcus) is a major cause of morbidity and mortality world-wide. Current vaccines target the pneumococcal capsule, and there are over 90 capsular serotypes. Serotyping pneumococcal isolates is therefore important for assessing the impact of vaccination programs and for epidemiological purposes. The World Health Organization has recommended latex agglutination as an alternative method to the &#8216;gold standard&#8217; Quellung test for serotyping pneumococci. Latex agglutination is a relatively simple, quick and inexpensive method; and is therefore suitable for resource-poor settings as well as laboratories with high-volume workloads. Latex agglutination reagents can be prepared in-house utilizing commercially-sourced antibodies that are passively attached to latex particles. This manuscript describes a method of production and quality control of latex agglutination reagents, and details a sequential testing approach which is time- and cost-effective. 
This method of production and quality control may also be suitable for other testing purposes.
