Karakterisering van de moleculaire mechanismen die de interacties van Salmonella onderbouwen met hun dierlijke gastheren wordt vooral bereikt door de studie van Salmonella enterica serotype Typhimurium (STM) in weefselkweek en diermodellen van infectie. Verkrijgen van inzicht in STM-gastheer interacties zal ons helpen begrijpen hoe Salmonella overleven en te groeien in gastheercellen. De eerste uitdaging in het bestuderen van deze interacties is om zoveel mogelijk deelnemende factoren identificeren van zowel de gastheer en pathogeen, maar deze pogingen zijn grotendeels geblokkeerd door de grote problemen van het omgaan met twee onafhankelijke complexe biologische systemen tegelijk, namelijk talrijke en Salmonella, onder fysiologische omstandigheden. Daarnaast is de grote repertOire van Salmonella en gastheer genen potentieel coderen factoren betrokken bij gastheer interacties vereisen high-throughput biologische platform om deze uitdaging aan te pakken. Computationele analyse van in vivo datasets geïdentificeerd 149 Salmonella genproducten die worden voorspeld om te interageren met ongeveer 300 gastheer genproducten (ongepubliceerde gegevens). De standaard Gentamicine bescherming assay niet de capaciteit van aantal mutanten efficiënt fenotype. Het volgende protocol is gedetailleerd en geïllustreerd om de gewijzigde Gentamicine bescherming assay met 96 putjes en het murine macrofaag cellijn RAW264.7 voeren. Vergeleken met het standaard protocol in 24-putjes, het gemodificeerde protocol heeft de volgende voordelen: 1) met 96 putjes kunnen tot 10 verschillende mutante stammen worden fenotype inclusief interne positieve en negatieve controles met voldoende statistische power;2) De variantie van de resultaten wordt aanzienlijk verminderd, omdat de mutante stammen worden getest in dezelfde kweekschaal en tegelijkertijd; 3) Het gebruik van meerkanaalspipetten verhoogt de doorvoersnelheid terwijl het verminderen van de vermoeidheid.
