Dans les années 1960, Bergström introduit une aiguille de biopsie percutanée similaire à celle décrite par Duchenne 4.2. Vingt ans après, Evans et al. Jour 5 la technique en appliquant une aspiration à travers le trocart de coupe de l&#39;aiguille Bergström. Cette modification peut améliorer le rendement de tissu de 3 à 5 fois 6,7 et est utilisé dans le cadre de recherches cliniques et biomédicales. Certains établissements d&#39;enseignement permettent une formation des chercheurs universitaires avec un doctorat, sous la supervision directe d&#39;un médecin, d&#39;obtenir des biopsies musculaires et d&#39;autres exigent un médecin conseil certifié pour effectuer la technique. L&#39;équipe de biopsie ASU a réalisé avec succès plus de 1600 biopsies musculaires au cours des 13 dernières années 8-15. Cette vidéo décrit la technique de biopsie percutanée à l&#39;aide d&#39;une aiguille Bergström modifié pour obtenir des échantillons de tissus squelettiques musculaire du muscle vaste externe de sujets humains. L&#39;aiguille est constituée d&#39;un Bergström canule extérieure avec une petite ouverture («fenêtre») sur le côté de la pointe et un trocart intérieur avec une lame de coupe à l&#39;extrémité distale. Dans des conditions d&#39;asepsie et d&#39;anesthésie locale, l&#39;aiguille est avancée dans le muscle squelettique par une incision dans la peau, le tissu sous-cutané, et le fascia. Ensuite, l&#39;aspiration est appliquée à la trocart interne, le trocart extérieur est tiré vers l&#39;arrière, le tissu musculaire squelettique est aspiré dans la fenêtre de la canule extérieure par l&#39;aspiration, et le trocart intérieur est rapidement fermé, coupe ou clipsage l&#39;échantillon de tissu musculaire squelettique ainsi . L&#39;aiguille est tourné de 90 °60, et une autre coupe. Ce processus peut être répété trois fois de plus. Cette technique de coupe multiple produit habituellement un échantillon de 100-200 mg ou plus chez des sujets sains et peut se faire immédiatement avant, pendant, et après une période d&#39;exercice ou d&#39;une autre intervention. Après pansement post-biopsie du site de l&#39;incision, les sujets reprennent généralement leurs activités de la vie quotidienne tout de suite et puissent participer pleinement à l&#39;activité physique vigoureuse dans les 48-72 heures. Les sujets doivent éviter lourde exercice de résistance pendant 48 heures pour réduire le risque de hernie du muscle à travers l&#39;incision dans le fascia.

La biopsie musculaire tel que décrit ci-dessus permet au chercheur de recueillir rapidement et de manière cohérente des échantillons de tissu du muscle squelettique. Le rendement typique chez des sujets sains de sport est de 200 mg ou plus en une seule passe avec 3-4 clips. La procédure prend 15 à 20 min, la majeure partie est consacrée à la préparation de l&#39;incision.
