Het zeewater werd gedecanteerd van elke centrifugebuis en elk monster werd vervolgens ondergedompeld biopsie, opgeslagen en in 4% paraformaldehyde getransporteerd. <p class=&quot;Jove_content&quot;&gt; SBFI imaging eerder is uitgevoerd op diverse biologische monsters, waaronder hele hersenen en het gehele hart menselijke weefsels intact muizenembryo&#39;s, zebravis embryo, en meerdere soorten dieren monsters met intacte botten 23-30. De meeste van deze studies gebruikt optische / lichtmicroscopie met ofwel fluorescentie of heldere veld technieken. Echter, zijn de studies uitgevoerd in ultra-hoge vergrotingen behulp van scanning electron serieblok gezicht beeldvorming in het verleden 31. In de onderhavige studie werd een gemodificeerd SBFI protocol ontwikkeld voor en toegepast op koralen voor de eerste keer. Omdat M. annularis en M. faveolata koraalpoliepen zijn 1-2 mm in dikte, zou geen van de routine lichtmicroscopie technieken kunnen doordringen van de gehele dikte van koraalpoliep weefsel. Daarom hebben we SBFI monstervoorbereiding protocol speciaal ontworpen voor koraal monsters. Daarnaast hebben we op maat ontworpen een stereomicroscoop houder, Die gemotoriseerd bewegen in zowel x en y richtingen. Deze inrichting neemt beelden van het blok van het monster dan verzamelen de gedeelten met bijvoorbeeld een microtoom voor de microscoop. We introduceerden ook een ander niet-lineaire optische twee-foton microscopische techniek op de foto dezelfde koraalpoliepen over de hele dikte van het koraal weefsel. Dit overwint de door SBFI gesteld wat ontkalking en de mogelijkheid van veranderingen in weefsel morfologie en volume (krimp) die kan worden geïnduceerd door monstervoorbereiding (dehydratie) en verwerkingsprotocollen beperkingen. Bovendien werden de emissie-profielen uit de koralen spectraal opgelost om hun piek uitstoot en variaties tussen de chromatoforen en de fotosynthetische zoöxanthellen identificeren. Deze resultaten werden in het kader van de methode en de individuele voordelen inzake acquisitie tijd, analysetijd en het vermogen om fijne structurele details lossen zonder afbreuk str geëvalueerductural integriteit van het koraal weefsel.

multimodal optical microscopy methods reveal polyp tissue morphology and structure in caribbean reef building corals
