角膜の問題は、世界中の生活の質を著しく低下させる何百万人もの人に影響を与える。角膜疾患は、無虹彩症またはスティーブンジョンソン症候群などの病気によって、ならびに、化学火傷または放射線などの外部要因によって引き起こされ得る。現在の治療は、（i）角膜移植片の使用及び（ii）幹細胞の使用は、研究室で展開され、キャリア（ 例えば 、羊膜）上で配信されます。これらの治療は、比較的成功しているが、残念ながら彼らは3-5年後に失敗することができます。詳細に幹細胞が角膜に常駐生理環境を模倣することができる新しい角膜生体材料デバイスを設計および製造する必要がある。輪部幹細胞は、フォークトのパリセーズとして知られている特定のニッチに輪部（角膜と強膜との間の円形領域）に位置しています。本研究では、2つの最先端の製造技術（マイクロ光とelectrospinnを組み合わせた新しいプラットフォーム技術を開発しましたある程度角膜縁を模倣角膜膜の製造のために）る。当社の膜は、フォークトのパリセーズが目に同じような保護を細胞に提供することを目指した人工micropocketsが含まれています。

エレクトロ微細加工リングは、マイクロ光エレクトロスピニング（ 図1および2）の組み合わせを用いて製造した。異なるサイズのPEGDA環はマイクロ光（ 図3）を使用して製造した。この技術は、cmのオーダーと微小特徴の同時取り込みを製作構造を可能にする。この場合には、350〜500ミクロンのmicropocketsを含む1.2〜1.6センチメートル範囲の直径のリングが（ 図4）を作製した。 製造殺菌および将来の臨床用途のための材料のパッケージングの観点からは、医療グレードの袋に真空パックが大幅PLGA膜（ 図5）の長期貯蔵を達成する能力を改善することが見出された。厚さ0.12mmの医療グレードのバッグ（PET /箔/ LDPE）の使用は、より長い貯蔵寿命を達成することができた。これはmembrを送信することによって調べたインドや膜における当社の共同研究者にanesは意図的に湿った条件（湿った培養器）で、室温で、37℃で、-20℃ヶ月間保存した。 図5は、での保存の意図的挑発的な条件を使用していることを示している37℃の湿った条件の下で、膜は非真空パックの条件で約1ヶ月間だけ安定していたが、真空パックされた条件（ 図5および表1）の下で3ヶ月保存を実現しました。 表1は、一つでも使用される袋の選択に注意を払った場合に水膨潤性の取り込みおよび繊維を助長するように選択条件下で達成することができる貯蔵条件の改善を実証する。 リングは、異なる条件での角膜縁の外植片からの細胞伸長（i）は6ヶ月（ 図6）のために保存作りたて及び（ii）のリングをリング支持した。細胞の移動は、4週間後に達成されたときのPL3D負傷モデルにPLGA膜をacing。細胞は、以前に剥皮角膜上で新しい上皮（ 図7）を作成した膜上に置かれた組織外植片から成長した。および陰性対照（負傷した角膜）正（無処理角膜）も、同じ時間の期間、培養で維持された。陰性対照は、任意の追加の細胞の不在下で新上皮の形成の欠如を確認した。免疫細胞は、彼らが角膜分化マーカーCK3（ 図7E）が陽性であったので、外植片から成長する細胞が角膜上皮細胞であることを示した。 <img alt="図1" fo:content-width="5in" src="/files/ftp_upload/51826/51826fig1highres.jpg" width="500" /> マイクロ光の図1の回路図は、PEGDAリングの作成 ​​のために設定します。 her.withinページの「常に &quot;=&gt;
