When performing CARS microscopic dissolution experiments there are a few critical aspects that need to be monitored during the experiment. Firstly, introducing the dissolution medium to the CARS flow cell causes the focus to move. This means that the image is immediately lost and it takes a few microns of objective adjustment to find the surface again. Secondly, there is risk of liquid leakage from the CARS flow cell if the glass cover breaks during the experiment. This can potentially cause liquid damage to the optics, so it is important to listen for any cracking sound that could mean the glass has broken. Finally, there is also a small chance that the piping can become blocked due to particulate matter in the system during the experiment, this can be seen as a sudden unusual change in the UV spectra and also through periodically checking the flow during the experiment.
Particulate blockage of the piping is mainly an issue with tablets that have been designed to disintegrate during dissolution. This is one of the limitations for this technique as this system requires the surface of the tablet to remain intact throughout the dissolution to allow imaging. In addition to disintegrating tablets, it is currently not possible to image tablets that are designed to swell during dissolution as this can lead to breakage of the CARS flow cell.
Imaging tablets during dissolution provides a greater understanding of what is occurring on the surface of a dissolving tablet. Conventional pharmaceutical dissolution methods focus only on the drug content dissolved in the dissolution medium which can identify whether the tablet passes or fails the required standard. However, in the case of a failed test it is difficult to determine what caused the failure. The case of a failed dissolution test is potentially where in situ dissolution analysis using CARS microscopy can provide answers.
Future applications for in situ dissolution analysis using CARS microscopy could include investigations using more complicated tablets containing more than one drug or excipient, in particular non-swelling sustained or controlled release dosage forms during formulation development. Additionally, it could be possible to investigate samples using biorelevant dissolution media creating conditions more closely related to in vivo. 
In conclusion, this work shows that CARS microscopy is capable of rapid chemically specific imaging based on Raman vibrational frequencies allowing selective imaging of the drug in a tablet containing both drug and excipient. Additionally, CARS microscopy combined with inline UV absorption spectroscopy is a powerful tool capable of monitoring the surface of tablets undergoing dissolution and correlating surface changes seen using CARS with changes in dissolution rate.

We hebben een nieuwe analytische methode om het oppervlak veranderingen op de tablet die een ontbinding met smalband CARS microscopie tijdens het opnemen van de opgeloste concentratie van het geneesmiddel met UV-absorptie spectroscopie tegelijk monitoren ontwikkeld. We illustreren het gebruik van deze methode beeldvorming tabletten die het modelgeneesmiddel theofylline gecombineerd met ethylcellulose ondergaan oplossen met water als oplosmedium.

coherent anti-stokes raman scattering (cars) microscopy visualizes pharmaceutical tablets during dissolution

Traditionele farmaceutische oplossingstesten bepalen de hoeveelheid geneesmiddel opgelost in de tijd door het meten geneesmiddelgehalte in het oplosmedium. Deze methode geeft weinig directe informatie over wat er gebeurt op het oppervlak van de oplossende tablet. Het oppervlak tablet samenstelling en structuur tijdens oplossen kan veranderen, is het essentieel om te volgen tijdens oplossingstesten. In dit werk coherent anti-Stokes Raman scattering microscopie wordt gebruikt om het imago van de oppervlakte van de tabletten tijdens het oplossen terwijl UV-absorptie spectroscopie wordt gelijktijdig verstrekken inline analyse van opgeloste concentratie van het geneesmiddel voor tabletten met een mengsel van 50% van theofylline anhydraat en ethyl cellulose. De metingen toonden aan dat in situ CARS microscopie kan beeldvorming selectief theofylline in aanwezigheid van ethylcellulose. Daarnaast is de theofylline anhydraat omgezet in theofylline monohydraat tijdens oplossen, met naaldvormige huilenStals groeien op het tablet oppervlak tijdens oplossen. De omzetting van theofylline anhydraat tot monohydraat, gecombineerd met verminderde blootstelling van het geneesmiddel aan het stromende oplosmedium resulteerde in verminderde oplossnelheden. Onze resultaten tonen aan dat in situ CARS microscopie gecombineerd met inline UV absorptie spectroscopie kan bewaken Farmaceutische tablet oplossen en correleren oppervlak verandert met veranderingen in de oplossnelheid.

In situ ontbinding analyse met behulp van CARS microscopie werd uitgevoerd op tabletten (12 mm diameter, plat oppervlak) met een 50:50 mengsel van het model geneesmiddel theofylline anhydraat en ethylcellulose met gedestilleerd water gepompt met 5 ml / min als de ontbinding medium. CARS beelden (512 x 512 pixels) werden verzameld om 1,12 sec bij de Raman trillingsfrequentie 2960 cm -1 die selectief is voor de theofylline inhoud van het tablet gedurende de duur van het experiment ontbinding. Figuur 2 toont geselecteerde frames uit de ontbinding video. Aan het begin van oplossing (figuur 2, tijdstip 0 sec) er gebieden van groene tonen de theofylline inhoud van het tablet en zijn ook donkere gebieden waar slechts ethylcellulose op het oppervlak van het tablet. In de donkere gebieden op het oppervlak van de tablet is het mogelijk om de ethylcellulose inhoud vaag bekijken. Dit is omdat het is gemeld dat ethyl cellulose heeft Raman trillingsfrequenties met maxima rond 2930 en 2975 cm-1 14. Na ongeveer 60 seconden blijkt er begin theofylline monohydraat kristalgroei op het oppervlak kan worden gezien als smalle naaldvormige kristallen naar buiten groeien van ten minste een kristal kern in het midden van het beeld (figuur 2 keer 60 sec) worden . Het monohydraat kristalgroei kan veel duidelijker gezien na 130 seconden (Figuur 2, tijd 130 sec). Bovendien, op tijdstip 130 sec blijkt dat het monohydraat kristal niet geheel is verspreid over het oppervlak van het tablet. Het lijkt alsof de aanwezigheid van de ethylcellulose regio fysiek geblokkeerd heeft de verlenging van het monohydraat naalden. Na 250 seconden, kan men zien dat het monohydraat dekking van het oppervlak is niet zo prominent dat suggereert dat de monohydraat kristallen zich beginnen te ontbinden. Figuur 2. De CARS video is beschikbaar als aanvullende informatie. Schaal bar is 50 micrometer. Ultraviolet (UV) spectroscopie is een vorm van absorptie spectroscopie met UV licht als de excitatiebron. UV-spectroscopie maatregelen elektron overgangen van de grondtoestand naar een aangeslagen toestand 15. Theofylline heeft een brede piek rond 270 nm terwijl ethylcellulose is vrijwel onoplosbaar in het oplosmedium wordt dus naar verwachting niet bijdragen aan het UV-spectrum opgenomen. Analyse van de dissolutie medium met de inline z-vormige UV-stroomcel kunnen we kwantitatief de hoeveelheid geneesmiddel opgelost tijdens oplossen. Figuur 3 toont de UV-oplossingsprofiel voor het oplossen van theofylline anhydraat met ethylcellulose tablet. De UV dissolutieprofiel (Figuur 3) laat zien dat de ontbinding van theofylline anhydraat begint het snel bereiken van een maximale concentratie van ongeveer 90 pg / ml binnen 120 seconden; na dit tijdstip de ontbinding tarief begint te dalen. De afname van de oplossnelheid kan door de aanwezigheid van de theofylline monohydraat (oplosbaarheid 6 mg / ml bij 25 ° C 16) kristallen op het oppervlak minder oplosbaar dan theofylline anhydraat (oplosbaarheid 12 mg / ml bij 25 ° C 16 zijn ) en duidelijk te zien in het CARS ontbinding video (figuur 2) op dit tijdstip. De geleidelijke verlaging dissolutiesnelheid kan ook gedeeltelijk worden verklaard bya vermindering theofylline blootstelling aan het stromende medium.
