Un procedimiento quirúrgico aquí descrito permite la observación directa de la dinámica celular mieloides dentro de los tumores intestinales en ratones vivos mediante el uso de ratones transgénicos reportero fluorescente y trazadores inyectables o anticuerpos. Para este propósito, una de cuatro colores, multi-región, disco giratorio microscopio confocal micro-lensed que permite formación de imágenes continua a largo plazo con una rápida adquisición de la imagen se ha utilizado. APC Min / + ratones que desarrollan múltiples adenomas en el intestino delgado se cruzan con los ratones c-FMS-EGFP para visualizar las células mieloides y con ratones ACTB-ECFPpara visualizar las células epiteliales intestinales de las criptas. También se describen procedimientos para el etiquetado de diferentes componentes tumorales, tales como vasos sanguíneos y neutrófilos, y el procedimiento para el posicionamiento del tumor para formación de imágenes a través de la superficie serosa. Time-lapse películas recopilados de varias horas de proyección de imagen permiten el análisis del comportamiento de las células mieloides in situ en el microambiente intestinal.
