SPR is een zeer gevoelige methode om moleculaire interacties te bestuderen, en is vaak de enige aanpak die real-time monitoring van voorbijgaande aard (nog belangrijker) interacties zorgt. Een voorbeeld is de transiënte interactie hierin gepresenteerde, die niet kan worden gedetecteerd door een andere methode (pull-down assays liposomen sedimentatie 6 assays). Bovendien, terwijl andere werkwijzen zijn beperkt tot metingen evenwicht (kwantitatief of kwalitatief), SPR is een van de weinige technieken die meten...

Eiwit-eiwit interacties (PPI); de vorming en dissociatie van eiwitcomplexen, zijn belangrijke gebeurtenissen in veel biologische processen (bijvoorbeeld, replicatie, transcriptie, translatie, signalering, cel-cel communicatie). Semi-kwantitatieve studies van de PPI worden vaak uitgevoerd met behulp van pull-down of immuno-neerslag experimenten. Echter, deze (en dergelijke) technieken beperkt het bereik van affiniteiten die meetbaar (lage micromolaire affiniteit en hoger) door de wasstappen die inherent zijn aan dergelijke technieken. Dergelijke &quot;eindpunt&quot; technieken kunnen niet van voorbijgaande of lage affiniteit interacties die vaak van grote biologische gevolgen zijn te identificeren. Bovendien wordt de temporele resolutie van deze aanpak uiterst beperkt, gewoonlijk orden van grootte lager dan de tarieven van de reactie. SPR overwint deze tekortkomingen te wijten aan de verhoogde gevoeligheid en superieure temporale resolutie van 1,2. SPR is een label vrij methode, en als zodanigmoleculaire interactie kan worden gemeten (als de massa veranderingen kunnen worden gedetecteerd). Naast PPI, is uitgebreid gebruikt om eiwit-ligand, eiwit-geneesmiddel (of klein molecuul), eiwit-DNA en eiwit-RNA-interacties 3-5 karakteriseren. De resultaten worden geregistreerd en uitgezet in real time, die een snelle gewijzigde experimentele omstandigheden en flexibel experimenteel ontwerp maakt. Het fysieke principe SPR gebaseerde instrumenten is het gebruik van een optisch systeem dat een verandering van de brekingsindex gemeten aan het sensoroppervlak op massaveranderingen 2. Eén van de interagerende partners (hierna ligand) geïmmobiliseerd op een polymeermatrix chip en de tweede molecule (hierna analyt) stroomt over het oppervlak. Analyt binding aan het ligand verandert de massa op het chip oppervlak. Deze massa verandering is direct en proportioneel verband met veranderingen in de brekingsindex van het oppervlak. De resultaten zijn uitgezet in realtimegepresenteerd als respons eenheden (RU) als functie van de tijd. Een dergelijke grafiek wordt aangeduid als Sensogram (bijvoorbeeld figuur 1). Aangezien SPR volgt de volledige tijdsverloop van de interactie, worden pre-evenwicht kinetische snelheidsconstanten verkregen, naast affiniteiten evenwicht. Zoals hieronder uiteengezet, de pre-evenwicht gedrag van een bepaald systeem bevat veel informatie, en geeft een ander perspectief dan evenwicht alleen. Zodra het systeem is gekalibreerd, experimenten zeer snel en vereist slechts kleine hoeveelheden eiwitmateriaal (microgram bedragen nanogram). Verzamelen de volledige kinetische informatie van een bepaald systeem kan op dagen. De hoge gevoeligheid van SPR biedt metingen die niet mogelijk zijn met behulp van een andere techniek 6. Echter, deze hoge gevoeligheid is een &#39;tweesnijdend zwaard&#39;, omdat het een belangrijke bron voor vals positieve data kan zijn. Elke factor die de brekingsindex van invloed is geregistreerd en uitgezet op de Sensogram. Als zodanig appropriate controles moeten worden gebruikt om vals-positieven te elimineren, en de steun van complementaire aanpak wordt sterk geadviseerd. Figuur 1 toont het verloop van een SPR-experiment met een NTA beklede sensorchip. De Sensogram in panel A geeft de injectie van de nikkel-ionen over de NTA-matrix (unsubtracted gegevens), en panelen BD de gegevens weer te geven na aftrek van het signaal afkomstig van de negatieve controle cel. Rode en blauwe pijlen geven injectie begin en einde, respectievelijk. Immobilisatie van de ligand op de chip geleidelijk verandert de massa tot injectie beëindigd. De stabiele plateau waargenomen na beëindiging van de injectie ligand weerspiegelt de stabiele associatie van de ligand aan het oppervlak (Figuur 1B, cyclus 2). Zodra een stabiele basislijn wordt bereikt een buffer wordt geïnjecteerd over het ligand en de celverwijzing (Figuur 1B, cyclus 3) .Deze injectie dient als een &#39;blanco-controle &quot;, en zal zijnafgetrokken tijdens de analyse. Bij inspuiting zijn kleine wijzigingen opgenomen, hetgeen een stroom van massa door de chip. Vervolgens in een aparte cyclus (Figuur 1B, cyclus 4), de analyt wordt geïnjecteerd; de geleidelijke toename RU vertegenwoordigt vereniging van de analyt aan het ligand. De bindingsplaatsen raken geleidelijk bezet tot evenwicht is bereikt. Zodra injectie eindigt, een daling van de SO&#39;s weerspiegelt dissociatie van het complex. Uit zulke sensogrammen pre-evenwicht en evenwicht snelheidsconstanten worden afgeleid (zie later). Figuur 1 toont een transiënte interactie tussen het ligand en de analyt. Wanneer de interactie is stabieler, de daling van de RU-niveau is langzamer, als gevolg van een lagere k d. Hierin beschrijven we een SPR experiment gericht op het karakteriseren van de interactie tussen een membraan transporter (detergens oplosbaar) en de functionele partner zijn verwante substraat bindend eiwit 6,7. De gekozen model-systeem is een ATP-binding cassette (ABC) transporter, ModBC-A van Archeaglobus fulgidus. Dit systeem is &#39;aan / uit&#39; tarieven geselecteerd voor zijn zeer reproduceerbare resultaten, hoge signaal-ruisverhouding en klassieke. Daarnaast homologen ABC transporters zijn beschikbaar om als belangrijke negatieve controles dienen. De vervoerder, ModBC (ligand A) wordt gewonnen uit het membraan met afwasmiddel, gezuiverd en geïmmobiliseerd op de chip. De oplosbare interactie partner, moda, is de analyt. Als negatieve controle ligand, wordt een andere ABC transporter RbsBC gebruikt (&quot;ligand B&quot;).

real time measurements of membrane protein:receptor interactions using surface plasmon resonance (spr)

Protein-protein interactions are pivotal to most, if not all, physiological processes, and understanding the nature of such interactions is a central step in biological research. Surface Plasmon Resonance (SPR) is a sensitive detection technique for label-free study of bio-molecular interactions in real time. In a typical SPR experiment, one component (usually a protein, termed 'ligand') is immobilized onto a sensor chip surface, while the other (the 'analyte') is free in solution and is injected over the surface. Association and dissociation of the analyte from the ligand are measured and plotted in real time on a graph called a sensogram, from which pre-equilibrium and equilibrium data is derived. Being label-free, consuming low amounts of material, and providing pre-equilibrium kinetic data, often makes SPR the method of choice when studying dynamics of protein interactions. However, one has to keep in mind that due to the method's high sensitivity, the data obtained needs to be carefully analyzed, and supported by other biochemical methods. 
SPR is particularly suitable for studying membrane proteins since it consumes small amounts of purified material, and is compatible with lipids and detergents. This protocol describes an SPR experiment characterizing the kinetic properties of the interaction between a membrane protein (an ABC transporter) and a soluble protein (the transporter's cognate substrate binding protein).

In het hier beschreven systeem wordt een NiNTA chip gebruikt om de His-tag membraan transporter 6,7 immobiliseren. Omdat het een homo-dimeer, wordt elke transporter dubbel gelabeld, verbetering van de binding aan de NiNTA chip.
