Chronische Lymfatische Leukemie (CLL), de meest voorkomende leukemie bij volwassenen in de westerse landen, is afgeleid van de accumulatie van monoklonale neoplastische CD5 + B-lymfocyten en is klinisch zeer heterogeen. Het kan als een pre-leukemische vorm, gedefinieerd als monoklonale B-cel CLL (MBL) 1,2,3 zijn. In andere gevallen kan de ziekte voorkomen, ofwel met een indolent beloop, kan dat jarenlang stabiel blijven voordat behandeling nodig hebben, of als een progressieve chemorefractory ziekte met sombere prognose ondanks therapie. Ten slotte kan vooruitgang in een vaak dodelijke hooggradige lymfomen (Richter syndroom-RS) 2,3,4. Patiënten worden meestal ingedeeld in twee subgroepen: goede en slechte prognose, gebaseerd op een reeks van prognostische factoren die aanvullende informatie over voorspellers van de ziekte uitkomst en overleving biedt. from a 2de-gel spot to protein function: lesson learned from hs1 in chronic lymphocytic leukemia

De identificatie van moleculen betrokken bij tumor initiatie en progressie is fundamenteel voor de biologie inzicht ziekte en, bijgevolg voor de klinische behandeling van patiënten. In dit werk zullen we een geoptimaliseerde proteomics aanpak voor de identificatie van moleculen die betrokken zijn bij de progressie van chronische lymfatische leukemie (CLL) te beschrijven. In detail zijn leukemische cellysaten gescheiden door 2-dimensionale elektroforese (2DE) en gevisualiseerd als &quot;spots&quot; op de 2DE gels. Vergelijkende analyse van proteomics kaarten maakt de identificatie van differentieel tot expressie gebrachte eiwitten (in termen van overvloed en post-translationele modificaties) die worden geplukt, geïsoleerd en geïdentificeerd door massaspectrometrie (MS). De biologische functie van de geïdentificeerde kandidaten worden getest door verschillende assays (bijvoorbeeld migratie, adhesie en F-actine polymerisatie), die we hebben geoptimaliseerd voor primaire leukemische cellen.

We geïsoleerd primaire leukemische B-cellen vormen PB van CLL-patiënten en analyseerden we proteomics kaarten. Monsters (n = 104) werden gegroepeerd in twee subsets op basis van de klinische kenmerken van elke patiënt (slechte prognose vs goede prognose) en vergelijkend onderzoek van 2DE-gel werd uitgevoerd.
