Arvelige eller erhvervede genetiske sygdomme kan føre til utilstrækkelig og dysfunktionelle syntese af proteiner og forårsage alvorlige sygdomme. Således er en ny terapeutisk fremgangsmåde til at inducere produktion af manglende eller defekte proteiner er det exogene levering af syntetisk modificeret mRNA i celler, som koder for det ønskede protein. Den exogene levering af kodning syntetisk messenger RNA (mRNA) for induktion af proteinsyntese i ønskede celler har et enormt potentiale inden for regenerativ medicin, grundlæggende cellebiologi, behandling af sygdomme og omprogrammering af celler. Her beskriver vi en trinvis protokol for generering af modificeret mRNA med nedsat immun aktivering potentiale og øget stabilitet, kvalitetskontrol af produceret mRNA, transfektion af celler med mRNA og verifikation af den inducerede protein-ekspression ved flowcytometri.
