De aandachtsproblemen set verschuiven taak (AST) werd ontwikkeld als een maatregel van aandacht en cognitieve flexibiliteit bij ratten meer dan tien jaar geleden 1,2. AST is gemodelleerd naar de intradimensional / extradimensional component van de Cambridge neuropsychologische Automated Accu (Cantab) wordt gebruikt om cognitieve disfunctie bij mensen en niet-humane primaten 3,4 identificeren. Terwijl de mogelijkheid om eenvoudige regels leren blijft intact, tekorten in het leren van een reactie wanneer de regels veranderd worden gevonden bij patiënten met diverse neuropsychiatrische stoornissen (bijvoorbeeld, schizofrenie, obsessieve compulsieve stoornis, depressie), neurodegeneratieve aandoeningen wijzigen (dwz ziekte van Parkinson) en bij patiënten met laesies aan de prefrontale cortex 5. Meer in het algemeen zijn deze patiënten beschreven met tekorten in cognitieve flexibiliteit. Analoge tekorten aangetoond bij zowel niet-humane primaten en knaagdieren wanneer laesies to de prefrontale cortex zijn geïnduceerd 1,6-8. Deze tekorten wijzen op een toestand van cognitieve inflexibiliteit of een verminderd vermogen om aandachtsproblemen set verschuiven. Een aandachtsproblemen set wordt gevormd wanneer een onderwerp leert dat een set van regels kunnen worden toegepast op complexe stimuli om relevant te onderscheiden van irrelevante cues. Bijvoorbeeld, in de AST zullen dieren leren letten en reageren op de desbetreffende cue (dwz graven gemiddeld) en negeert een irrelevante cue (dwz geur), door het koppelen van een voedselbeloning met het medium. Deze vereniging wordt dan versterkt in de volgende taken waar de soort graven medium en geur verandert, maar de gepaarde associatie tussen medium en beloning blijft. Dit versterkt regel vormt een cognitieve set. Twee etappes in de AST-protocol maatregel aspecten van cognitieve flexibiliteit: omkering en de extra-dimensionale verschuiving. Bij de omkering stadium, de eerder negatieve prikkels binnen één Dimension (gemiddeld in dit voorbeeld) is nu positief dat het dier betwist de positieve prikkels uit de vorige fase negeren. Als bijvoorbeeld vilt graven medium was positief stimuli in de voorgaande fase en papier was de negatieve stimuli, nu het omgekeerde waar. Dit daagt de flexibiliteit van het dier in dat het de aandachtsprocessen set moet doen (dwz, medium is de relevante dimensie), terwijl het veranderen van de regel geleerd voor stimulans en beloning pair binnen een dimensie. De vorming van een aandachtsbias set wordt uitgedaagd op de extra-dimensionale verschuiving stadium, wanneer de irrelevante dimensie (geur in dit voorbeeld) wordt de betreffende dimensie. Een perseverative respons, zoals aangegeven door een voortdurende keuze met de eerder geleerde regel aan beide stadium weerspiegelt een tekort in cognitieve flexibiliteit. Laesie studies in zowel niet-humane primaten en knaagdieren hebben aangetoond specifieke gebieden van de prefrontale cortex kan worden toegeschreven aan het vermogen om perfORM bepaalde stadia van de AST 1,9. Letsels aan de orbitofrontale cortex (OFC), een subgebied van de prefrontale cortex (PFC), is aangetoond dat tekorten omkering leren op de AST 8 induceren. Bovendien, letsels aan de mediale prefrontale cortex (MPFC) leiden tot specifieke problemen bij het uitvoeren van de extra-dimensionale verschuiving. Terwijl talrijke studies op de AST werden uitgevoerd bij ratten en niet-menselijke primaten, hebben betrekkelijk weinig AST gebruikt als maat voor cognitieve functie bij muizen. Gezien het gemak van genetische manipulatie bij muizen, en de kritische noodzaak om prefrontale cortex functie te meten in het bestuderen van diverse aandoeningen, aanpassen en valideren van deze gedragsproblemen maatregel bij muizen is een belangrijke aanvulling op het onderzoek naar ziekten die samenhangen met PFC disfunctie. De hier beschreven protocol is een modificatie van de procedure Birrell en Brown 1 die is geoptimaliseerd voor toepassing in muizen en weerspiegelt veel van de muis specific aanpassingen die eerder zijn gemeld 2,6,10-13.

the attentional set shifting task: a measure of cognitive flexibility in mice

Cognitive impairment, particularly involving dysfunction of circuitry within the prefrontal cortex (PFC), represents a core feature of many neuropsychiatric and neurodevelopmental disorders, including depression, post-traumatic stress disorder, schizophrenia and autism spectrum disorder. Deficits in cognitive function also represent the most difficult symptom domain&#160;to successfully treat, as serotonin reuptake inhibitors and tricyclic antidepressants have only modest effects. Functional neuroimaging studies and postmortem analysis of human brain tissue implicate the PFC as being a primary region of dysregulation in patients with these disorders. However, preclinical behavioral assays used to assess these deficits in mouse models which can be readily manipulated genetically and could provide the basis for studies of new treatment avenues have been underutilized. Here we describe the adaptation of a behavioral assay, the attentional set shifting task (AST), to be performed in mice to assess prefrontal cortex mediated cognitive deficits. The neural circuits underlying behavior during the AST are highly conserved across humans, nonhuman primates and rodents, providing excellent face, construct and predictive validity.
