Den uppmärksamhets set skiftande uppdrag (AST) utvecklades som ett mått på uppmärksamhet och kognitiv flexibilitet hos råttor över ett decennium sedan 1,2. AST är modellerad efter intradimensional / extradimensional komponent i Cambridge Neuropsychological Test Automated Batteri (Cantab) som används för att identifiera kognitiv dysfunktion i människor och icke-mänskliga primater 3,4. Även om förmågan att lära sig enkla regler förblir intakt, att underskott i lärandet ändra ett svar när reglerna har ändrats påträffas hos patienter som lider av en mängd olika neuropsykiatriska störningar (dvs, schizofreni, tvångssyndrom, depression), neurodegenerativa sjukdomar (dvs, Parkinsons sjukdom) och hos patienter med lesioner till pre-frontala cortex 5. Mer allmänt är dessa patienter beskrivs ha underskott i kognitiv flexibilitet. Analoga underskott har visats i både icke-mänskliga primater och gnagare lesioner to prefrontala cortex har inducerats 1,6-8. Dessa underskott indikerar ett tillstånd av kognitiv stelbenthet eller en nedsatt förmåga att skifta uppmärksamhets set. En uppmärksamhets set bildas när ett ämne får reda på att en uppsättning regler kan tillämpas på komplexa stimuli för att skilja relevanta ur irrelevanta ledtrådar. Till exempel, i AST, kommer djuren lära sig att uppmärksamma och svara på den relevanta cue (dvs., grävning medium) och ignorera en irrelevant cue (dvs., lukt), genom att para en matbelöning med mediet. Denna förening sedan förstärks i efterföljande uppgifter där typen av grävning medel och luktförändringar, men den parade association mellan medium och belöning kvar. Denna förstärkta regel bildar en kognitiv set. Två faser inom AST protokoll mäter aspekter av kognitiv flexibilitet: återföring och den extra-dimensionella skift. Vid återföring stadiet, tidigare negativa stimuli inom en Dimension (medium i detta exempel) är nu positivt, vilket utmanar djuret att ignorera de positiva stimuli från föregående steg. Till exempel, om kändes gräva mediet var de positiva stimuli i föregående steg och papper var de negativa stimuli, nu det omvända är sant. Detta utmanar djurets flexibilitet i att den är bunden av sitt uppmärksamhets set (dvs, är den relevanta dimensionen mediet) samtidigt förändra regeln omar för stimulans och belöning paret inom en dimension. Bildandet av en uppmärksamhets set utmanas vid extradimensionell skift skede då irrelevant dimensionen (lukt i detta exempel) blir aktuella dimensionen. En perseverative respons, vilket indikeras av en fortsatt val med hjälp av tidigare lärt regeln, antingen vid scenen speglar ett underskott i kognitiv flexibilitet. Lesioner till orbitofrontal cortex (OFC), en underregion av prefrontala cortex (PFC), har visat sig inducera underskott i återföring lärande på AST 8. Även ett stort antal studier som använder AST har utförts på råtta och primater, har relativt få utnyttjat AST som ett mått på kognitiv funktion hos möss. Med tanke på den enkla genetisk manipulation i möss, och den kritiska behovet av att mäta prefrontala cortex funktion i att studera en mängd olika sjukdomar, anpassa och validera denna beteende åtgärd i möss är ett viktigt tillskott till forskningen om sjukdomar som förknippas med PFC dysfunktion. Protokollet beskrivs här är en modifiering av förfarandet genom Birrell och Brown 1 som är optimerad för användning i möss och återspeglar många av mus-specific anpassningar som tidigare har rapporterats 2,6,10-13.

the attentional set shifting task: a measure of cognitive flexibility in mice

Cognitive impairment, particularly involving dysfunction of circuitry within the prefrontal cortex (PFC), represents a core feature of many neuropsychiatric and neurodevelopmental disorders, including depression, post-traumatic stress disorder, schizophrenia and autism spectrum disorder. Deficits in cognitive function also represent the most difficult symptom domain&#160;to successfully treat, as serotonin reuptake inhibitors and tricyclic antidepressants have only modest effects. Functional neuroimaging studies and postmortem analysis of human brain tissue implicate the PFC as being a primary region of dysregulation in patients with these disorders. However, preclinical behavioral assays used to assess these deficits in mouse models which can be readily manipulated genetically and could provide the basis for studies of new treatment avenues have been underutilized. Here we describe the adaptation of a behavioral assay, the attentional set shifting task (AST), to be performed in mice to assess prefrontal cortex mediated cognitive deficits. The neural circuits underlying behavior during the AST are highly conserved across humans, nonhuman primates and rodents, providing excellent face, construct and predictive validity.
