Det overordnede mål for denne video var at vise en real-time overvågning procedure for at erhverve systemiske og hepatiske hæmodynamiske parametre. Begrundelsen for at udvikle denne fremgangsmåde er dens store værdi for eksperimentelle interventioner i mus, der kræver at få systemiske og hepatiske hæmodynamiske parametre. Proceduren kan anvendes til naive dyr og under eller efter en given hepatobiliær eksperimentel kirurgisk intervention, såsom delvis hepatektomi, portal vene ligering og levertransplantation. Erhvervelse af hepatiske hæmodynamiske data i gnavere kræver den foreslåede invasiv procedure. Nedsat perfusion kan ikke opnås non-invasivt. Men der er alternativer til erhvervelse af det systemiske blodtryk. Overvågning teknikker såsom halemanchetmetoden teknik 8 er blevet anvendt til at erhverve blodtrykket hos både rotter og mus. Halen manchet teknik kan anvendes i consciskellige dyr. Ved måling af blodtryk, dyret skal placeres og fikseres i en bestemt ubehagelig position. I den manuelle af halen-manchet-enhed, producenten, at mus kan blive nervøs og stresset, som kan mindske omsætning i halen. Under denne omstændighed, kan den perifere blodtryk erhvervet i halen være meget lavere end den centrale blodtryk. Vi har allerede rapporteret om en lignende intraoperativ overvågningsprocedure for rotter resulterer i en omfattende serie af fysiologiske hæmodynamiske data er sammenlignelige med single indberettede data fra andre grupper 7. Derfor betragtede vi denne fremgangsmåde til at repræsentere et godt grundlag for at tilpasse den til musen, en art 10 gange mindre end rotter. Den vigtigste forskel i forhold til rotte-procedure er anvendelsen af ​​Millar kateter for at erhverve blodtryk data i stedet for en væskebaseret katetersystem. Strømdata blev også erhverves med transsoniske flow sonder, bare langt mindre end for de tilsvarende rotte fartøjer. På grund af den lille størrelse af dyret, instrumentering af mus er teknisk udfordrende, men realistisk. Når instrumentering er afsluttet, datafangst og primær liv dataanalyse er enkel, da kan anvendes en foruddefineret indstilling fil. Indstillingen fil skal defineres en gang ved begyndelsen af ​​en række eksperimenter og kan lagres og anvendes til alle efterfølgende eksperimenter. Indtil nu har vi anvendt denne procedure til vurdering hepatiske hæmodynamiske effekter i akutte forsøg. Vi målte CAP og PVP før og umiddelbart efter 70% delvis hepatektomi (PH) og fastspænding / de-fastspænding eksperimenter. Vi fastspændt lever-duodenale ledbånd i højre lap repræsenterer 20% af levermasse efterfulgt af kort (5 min) fastspænding af medianen og venstre laterale lap repræsenterer totalt 90% af leveren masse. De-fastspænding startede med at frigive klemmen fra højre lap efterfulgt af frigøre median og venstre laterale lap. Maksimal fastspænding tid var under 10 minutter.

Brugen af ​​musemodeller i eksperimentel forskning er af enorm betydning for studiet af hepatisk fysiologi og patofysiologi. Men på grund af den lille størrelse af musen, tekniske detaljer ved intraoperativ kontrolprocedure egnet til mus sjældent beskrevet. Vi har tidligere rapporteret om en kontrolprocedure for at opnå hæmodynamiske parametre for rotter. Nu har vi tilpasset fremgangsmåde til at erhverve systemiske og hepatiske hæmodynamiske parametre i mus, en art ti gange mindre end rotter. Denne film viser instrumentering af dyrene samt datafangst proces er nødvendig for at vurdere systemiske og hepatiske hæmodynamik hos mus. Vitale parametre, herunder kropstemperatur, respirationsfrekvens og puls blev registreret under hele proceduren. Systemiske og hepatiske hæmodynamiske parametre forblev inden for normalområdet under denne procedure. Denne procedure er udfordrende, men realistisk. Vi har allerede anvendt denne procedure til vurdering hepatisk hæmodynamik hos normale mus såvel som under 70% partiel hepatektomi og leverlap fastspænding eksperimenter. Mean PVP efter resektion (n = 20) var 11,41 ± 2,94 cmH 2 O, der var signifikant højere (P &lt;0,05) end før resektion (6,87 ± 2,39 cmH 2 O). Resultaterne af leverlap fastspænding eksperiment viste, at denne kontrolprocedure er følsom og egner sig til påvisning små ændringer i portalen pres og portal flow.
