efficient ips cell generation from blood using episomes and hdac inhibitors

כתב יד זה מדגים פרוטוקול ליעילות יצירת תאים ללא אינטגרציה אנושית הנגרם גזע pluripotent (iPSCs) מהדם היקפי באמצעות פלסמידים episomal וdeacetylase היסטון מעכבים (HDAC). היתרונות של גישה זו כוללים: (1) השימוש בכמות מינימאלית של דם היקפי כחומר מקור; (2) nonintegrating וקטורי תכנות מחדש; שיטה חסכונית ליצירת iPSCs החופשי וקטור (3); transfection יחיד (4); ו- (5) השימוש במולקולות קטנות כדי להקל על תכנות מחדש אפיגנטיים. בקצרה, תאים היקפיים mononuclear הדם (PBMCs) מבודדים מדגימות לקיחת דם שגרתיות ולאחר מכן בתרבית גורמי גדילה מוגדרת להניב אוכלוסיית תא אב כדורית אדומה שגשוג מאוד כי להפליא ניתן לתכנות מחדש.
