efficient ips cell generation from blood using episomes and hdac inhibitors

Dette manuskriptet illustrerer en protokoll for effektivt å skape integrasjon frie menneskeskapte pluripotent stamceller (iPSCs) fra perifert blod ved hjelp episomale plasmider og histondeacetylase (HDAC) hemmere. Fordelene med denne tilnærming omfatter: (1) bruk av en minimal mengde av perifert blod som en kilde materiale; (2) nonintegrating omprogrammering vektorer; (3) en kostnadseffektiv fremgangsmåte for å generere vektor frie iPSCs; (4) en enkelt transfeksjon; og (5) anvendelse av små molekyler for å lette epigenetisk omprogrammering. I korthet, blir perifere mononukleære blodceller (PBMC) isolert fra flebotomi rutineprøver og deretter dyrket i definerte vekstfaktorer for å gi et sterkt proliferativ erytrocytt progenitor-cellepopulasjonen som er bemerkelsesverdig mottagelig for omprogrammering. Nonintegrating, nontransmissible episomale plasmider uttrykker OCT4, SOX2, KLF4, MYCL, LIN28A, og en p53 kort hårnål (sh) RNA er introduced inn i de avledet erytroblaster via en enkelt nucleofection. Kotransfeksjon av en episom som uttrykker forbedret grønt fluorescerende protein (EGFP) gir mulighet for enkel identifisering av transfekterte celler. En separat replikasjon-manglende plasmid som uttrykker Epstein-Barr-atom antigen 1 (EBNA1) blir også tilsatt til reaksjonsblandingen for økt ekspresjon av episomale proteiner. Transfekterte celler blir deretter sådd ut på et lag av bestrålte mus embryoniske fibroblaster (iMEFs) for fortsatt omprogrammering. Så snart IPSC-kolonier som vist på omtrent tolv dager etter nucleofection, er HDAC-inhibitorer tilsettes til mediet for å lette epigenetisk remodeling. Vi har funnet at inkludering av HDAC-inhibitorer øker rutinemessig generering av fullt omprogrammeres IPSC kolonier av 2-fold. Når IPSC kolonier utviser typisk humane embryonale stamceller (hESC) morfologi, de er forsiktig overført til individuelle iMEF-belagt vevskulturplater for fortsatt vekst og ekspansjon.

Tre dager etter nucleofection og før utplating av cellene på nucleofected iMEFs, bør effektiviteten av vellykket nucleofection estimeres ved fluorescens mikroskopi for EGFP. Figur 1 viser et typisk eksperiment nucleofection med ca. 5-10% av den totale cellepopulasjon som uttrykker EGFP. Omprogrammerte IPSC kolonier vil begynne å dukke opp ca to uker etter nucleofection.
