De generatie van hartspiercellen uit hPSCs, inclusief hESC en hiPSC, kan functioneren als een in vitro model van de zeer vroege menselijke cardiale ontwikkelingsprocessen, die inzicht geven in etappes anders niet toegankelijk zijn voor mechanistische studies. Dit model biedt unieke mogelijkheden om de moleculaire pathways die cardiale lineage inzet en lot van de cel specificatie controle te bestuderen. De laatste jaren heeft de mogelijkheid om efficiënt genereren cardiomyogene cellen van hPSCs sterk verbeterd 1-15. Echter, onder protocollen er cellijn variatie met betrekking tot de efficiëntie van het genereren cardiomyogene cellen en timing waarmee de cellen tot expressie kamer merkers (bijvoorbeeld ventrikel en atria). Idealiter voor toekomstige toepassingen van dit modelsysteem homogenere populatie van functioneel gedefinieerde cellen gewenst. Kortom, de voordelen van dit protocol vergeleken met de vorige verslagen zijn onder meer de kosten-effectiveness, reproduceerbaarheid, en zijn hoge efficiency voor het genereren van hartspiercellen tussen meerdere HPSC lijnen, met inbegrip van hESC en hiPSC lijnen. Flow cytometrie is een krachtige analytische tool voor het beoordelen van de kwaliteit van de cellen in de cultuur en het bepalen subpopulatie homogeniteit, en met de juiste experimentele opzet, kan kwantitatieve metingen geven. Zoals met alle antilichamen gebaseerde strategieën, nauwkeurige interpretatie van experimentele resultaten vereist dat elementen van de test ontwerp met inbegrip van de concentratie en fixatie antilichaam en permeabilisatie voorwaarden (wanneer de doelgroep intracellulaire antigenen) worden zorgvuldig getest voor elk antilichaam als sub-optimale omstandigheden aanzienlijk efficiëntie van antilichaam invloed bindend, en dus, de interpretatie van de resultaten. De keuze van merkers, en hun overeenkomstige antilichamen worden gebruikt om de zuiverheid van in vitro cardiomyogenesis beoordelen varieert tussen rapporten. TNNT2 is beschouwd als een indicator van de cellen zich in voor de cardiomyogene lot en wordt routinematig gebruikt om de efficiëntie van de cardiale differentiatie protocollen te beoordelen. Er is een dringende noodzaak om de aanpak voor het repareren van het beschadigde hart, het ontdekken van nieuwe therapeutische geneesmiddelen die geen toxische effecten op het hart hebben, en het verbeteren van de strategieën om nauwkeurig te modelleren hart-en vaatziekten te ontwikkelen. Het potentieel van humane geïnduceerde pluripotente stamcellen (hiPSC) technologie benutten om de hartspier te genereren &quot;in een schotel&quot; voor deze toepassingen blijft hoog enthousiasme te genereren. De laatste jaren heeft de mogelijkheid om efficiënt genereren cardiomyogene cellen van humane pluripotente stamcellen (hPSCs) sterk verbeterd, biedt ons nieuwe mogelijkheden model zeer vroege stadia van menselijke hartontwikkeling zonder dergelijke. In tegenstelling tot vele eerdere werkwijzen hartspierdifferentiatie protocol beschreven here ofwel geen celaggregatie of toevoeging van activine A of BMP4 vereisen en robuust genereert celkweken die sterk positief zijn voor cardiaal troponine I en T (TNNI3, TNNT2), iroquois- klasse homeodomein eiwitIRX-4 (IRX4), myosine regulerende lichte keten 2, ventriculaire / hartspier isovorm (MLC2v) en myosine regulerende lichte keten 2, atriale isovorm (MLC2a) op dag 10 in alle menselijke embryonale stamcellen (hESC) en hiPSC lijnen getest datum. Cellen kunnen worden gepasseerd en onderhouden voor meer dan 90 dagen in de cultuur. De strategie is technisch eenvoudig te implementeren en kosteneffectief. Karakterisering van cardiomyocyten afgeleid van pluripotente cellen omvat vaak de analyse van referentiemarkeringen, zowel op mRNA en eiwitniveau. Voor eiwitanalyse flowcytometrie een krachtig analytisch hulpmiddel voor de beoordeling van cellen in kweek en het bepalen subpopulatie homogeniteit. Echter, technische variatie in monstervoorbereiding significante invloed kwaliteit van flowcytometrie data. Daarom moet standaardisatie van kleuringsprotocollen vergelijkingen tussen verschillende differentiatie strategieën vergemakkelijken. Bijgevolg geoptimaliseerd kleuringsprotocollen voor de analyse van IRX4, MLC2v, MLC2a, TNNI3 en TNNT2 door flowcytometrie beschreven.

Op dag 0, cellen 100% confluent met compact morfologie en minimale celresten.
