Under utveckling, ger en enda cell upphov till en hel organism i en mycket komplex process som involverar både celldelningar och rörelser. För att bättre förstå de biologiska principer dynamiken i cellens beteende, har utvecklingsbiologer börjat använda fluorescens-baserade in vivo avbildningstekniker. Specifika facken i celler, såsom cellmembran, kan antingen märkas av behandlingar med fluorescerande färger, ett tillvägagångssätt hämmas av brist på specificitet och vävnadspenetration 1, eller av den specifika införande i embryot av exogena mRNA kodar fluorescerande proteiner 2. Olika tekniker kan användas för effektiv leverans av exogena föreningar, såsom mRNA. Dessa innefattar, men är inte begränsade till, mikroinjektion, elektroporering, bombardemang med mikropartiklar, lipofektion och transduktion 3,4. Även om alla dessa metoder kan användas för att införa exogena föreningar till enväxande embryot, bara mikroinjektion tillåter tillämpningen av fördefinierade och exakta mängder i varje cell 3. Mikroinjektion tekniker har beskrivits för alla större utvecklingsmodellsystem 4 (t.ex. fruktflugor, nematod maskar, zebrafisk, grodor, möss) såväl som för några alternativa modeller 4, inklusive de som används för jämförande studier som syftar till att förstå utvecklingen av utvecklingsmekanismer (t.ex. havsanemoner, Annelid maskar, sjöborrar, ascidian manteldjur, den Lansettfiskar amphioxus). Cephalochordates, som tillsammans med manteldjur och ryggradsdjur fastställer chordate fylum, är särskilt väl lämpade modeller för att studera utvecklingen av chordates och diversifieringen av ryggradsdjur från en ryggradslös förfader 5-8. Den Lansettfiskar härstamning avvek mycket tidigt under chordate evolution; och ännu existerande cephalochordates, som indelas i tre släkten (Branchiostoma, Asymmetron och Epigonichthys), liknar ryggradsdjur både vad gäller övergripande anatomi och genomet arkitektur 5-8. Av de cirka 30 arter av cephalochordates som har beskrivits hittills, fem är tillgängliga för embryologiska och utvecklingsstudier 6,9: Asymmetron lucayanum (Bahama LANSETTFISK), Branchiostoma floridae (Florida amphioxus), Lansettfisk (den europeiska amphioxus), Branchiostoma belcheri (den kinesiska amphioxus) och Branchiostoma japonicum (den japanska amphioxus). Mogna vuxna i tre av dessa arter (B. lanceolatum, B. belcheri och B. japonicum) kan förmås att leka on-demand under häckningssäsongen 10,11. Dessutom, åtminstone för B. lanceolatum, kan effektiv lek också induceras i artificiellt havsvatten 12, vilket gör detta särskilt Lansettfiskar arter tillgänglig för labotalet som inte har tillgång till naturligt havsvatten. Kombinationen, i B. lanceolatum, en bekväm och pålitlig tillgång till embryon med en effektiv leveransmetod, såsom mikroinjektion, hittills den enda leverans teknik utvecklad i amphioxus (i både B. floridae och B. belcheri) 13-15, kommer att möjliggöra utvecklingen av en roman svit av manipulativa tekniker, bland annat härstamning tracing- och dynamisk cellbeteendebaserade strategier. Ett protokoll för effektiv mikroinjektion av mRNA för att uttrycka fluorescerande proteiner i B. lanceolatum embryo följaktligen utvecklats. Dessutom, för att ge en grundläggande verktygslåda för live avbildning av B. lanceolatum embryon undersöktes vektorsystem utvecklats som tillåter membranassocierat och nukleär expression av fluorescerande proteiner. För membraninriktning förhöjdes grönt fluorescerande protein (EGFP) fusionerad till den humana HRAS CAAX boxen och nukleär lokalisering av mCherry och EGFP varerhålls genom sammansmältning till zebrafisk histon 2B (H2B) exon (Figur 1, Kompletterande Arkiv 1). Dessutom med målet att optimera protein översättning, de Kozak-sekvenser och kodon av konstruktionerna har modifierats och anpassats till användning i B. lanceolatum. Sammantaget kommer den injektionsteknik och expressionsvektorer presenteras här tjäna som underlag för generering av nya experimentella metoder för cephalochordates, särskilt analyser använder den senaste fluorescens-baserade in vivo avbildningstekniker.

expression of fluorescent proteins in branchiostoma lanceolatum by mrna injection into unfertilized oocytes

We report here a robust and efficient protocol for the expression of fluorescent proteins after mRNA injection into unfertilized oocytes of the cephalochordate amphioxus, Branchiostoma lanceolatum. We use constructs for membrane and nuclear targeted mCherry and eGFP that have been modified to accommodate amphioxus codon usage and Kozak consensus sequences. We describe the type of injection needles to be used, the immobilization protocol for the unfertilized oocytes, and the overall injection set-up. This technique generates fluorescently labeled embryos, in which the dynamics of cell behaviors during early development can be analyzed using the latest in vivo imaging strategies. The development of a microinjection technique in this amphioxus species will allow live imaging analyses of cell behaviors in the embryo as well as gene-specific manipulations, including gene overexpression and knockdown. Altogether, this protocol will further consolidate the basal chordate amphioxus as an animal model for addressing questions related to the mechanisms of embryonic development and, more importantly, to their evolution.
