Små interfererende RNA (sirnas) er dobbelt-RNA-molekyler som kan undertrykke uttrykket av gener. I de senere årene har sirnas blitt utviklet som en ny generasjon av biodrugs som viser terapeutisk potensial for fremtidig bruk i kliniske applikasjoner 1-5. Men fortsatt er vellykket implementering av siRNA terapi en betydelig utfordring, på grunn av nedbrytning av nukleaser, dårlig intracellulære opptaket, lav transfeksjonseffektivitet og ineffektiv utgivelse fra endosomet / lysosomet 5. Mange av disse hindringer kan overvinnes ved utvikling av levering plattformer, som sikkert og effektivt kan levere siRNA til sykt vev. Sammenlignet med virusbærere, ikke-virale plattformer gir flere fordeler, for eksempel sikkerhet, lave kostnader og enkel skreddersøm. Spesielt kationiske nanopartikler, så som polymerer og lipider, har vist seg nyttig for siRNA levering 3. Tidligere har vi utviklet en skiveformede drug leveringssystem, betegnet flertrinns vektor (MSV). Denne plattformen er basert på sekvensielle trinn, hvor ett kjøretøy er frigjort fra hverandre. Det første trinn kjøretøy er en mikropartikkel laget av biologisk nedbrytbare porøse silisium (PSI), mens den andre fasen kjøretøyer er nanopartikler ladet med medikamenter eller kontrastmidler 6,7. Nanopartikler, som er innebygd i PSI materiale, er gradvis utgitt som Si degraderer 8. En fordel med å bruke Si partikler er at morfologi og overflateegenskaper kan enkelt tilpasses for å oppnå optimal biodistribusjon og narkotika utgivelse. Nylig ble den vellykkede bruk av MSV-plattform for levering av siRNA liposomer til tumorvevet vist i en ovarian og brystkreft musemodell 9, 10. I dette arbeidet har vi fabrikkert en universell levering system for siRNA basert på prinsippene i MSV-plattformen. Effekten av dette avgivelsessystemet har tidligere blitt demonstrert ossing forskjellig siRNA molekyler 11. Systemet er et polykation-funksjonalisert porøs silisium (PCPS) bærer, som består av PSI podet med polyetylenimin (PEI) og arginin (Arg). PEI kan hjelpe til å danne elektrostatiske interaksjoner med siRNA, mens Arg og PSI kan tjene til å redusere giftigheten av PEI, som tidligere demonstarted 11. I tillegg kan tilstedeværelsen av PEI bistå i intracellulært opptak og endosomale flukt, mens psi mikropartikler mulig siRNA beskyttelse og forlenget frigivelse. PSI partiklene gradvis nedbrytes under fysiologiske betingelser, for derved å resultere i dannelsen av Arg-PEI / siRNA nanopartikler (figur 1), som har en distinkt morfologi og en smal størrelsesfordeling 11. Dette PCPS plattformen skiller seg fra konvensjonell MSV, siden de andre scenenanopartikler ikke er i utgangspunktet present i transportøren, men dannes over tid som første-trinns bærer degraderer 11, 12. Den siRNA lasteeffektivitet, cytotoksisitet og genet stanse effektiviteten av PCPS systemet har blitt evaluert in vitro. Transfeksjonseffektiviteten ble målt ved hjelp av siRNA mot ataxia telangiectasia mutated (ATM) onkogen, som er involvert i DNA-reparasjon 10. porous silicon microparticles for delivery of sirna therapeutics

Small interfering RNA (siRNA) can be used to suppress gene expression, thereby providing a new avenue for the treatment of various diseases. However, the successful implementation of siRNA therapy requires the use of delivery platforms that can overcome the major challenges of siRNA delivery, such as enzymatic degradation, low intracellular uptake and lysosomal entrapment. Here, a protocol for the preparation and use of a biocompatible and effective siRNA delivery system is presented. This platform consists of polyethylenimine (PEI) and arginine (Arg)-grafted porous silicon microparticles, which can be loaded with siRNA by performing a simple mixing step. The silicon particles are gradually degraded over time, thereby triggering the formation of Arg-PEI/siRNA nanoparticles. This delivery vehicle provides a means for protecting and internalizing siRNA, without causing cytotoxicity. The major steps of polycation functionalization, particle characterization, and siRNA loading are outlined in detail. In addition, the procedures for determining particle uptake, cytotoxicity, and transfection efficacy are also described.
