Detta protokoll beskriver en metod för framgångsrik leverans och transfektion av siRNA in i celler. Små störande RNA (siRNA) är dubbelsträngade RNA-molekyler som kan undertrycka uttrycket av gener. Under de senaste åren har siRNA utvecklats som en ny generation biodrugs som visar terapeutisk potential för framtida användning i kliniska tillämpningar 1-5. Dock kvarstår ett framgångsrikt genomförande av siRNA terapi en betydande utmaning, på grund av nedbrytning av nukleaser, dålig intracellulär upptag, låg transfektionseffektivitet och ineffektiv frigivning från endosomen / lysosome 5. Många av dessa hinder kan övervinnas genom utveckling av leveransplattformar, vilket säkert och effektivt kan leverera siRNA till sjuk vävnad. Jämfört med virusbärare, icke-virala plattformar ger flera fördelar, såsom säkerhet, låg kostnad och enkel skrädderi. I synnerhet katjoniska nanopartiklar, såsom polymerer och lipider, har visat sig vara användbart för siRNA leverans 3. Tidigare har vi tagit fram en skivformig drug leveranssystem, kallade flerstegs vektorn (MSV). Denna plattform är baserad på varandra följande steg, i vilket ett fordon är frigjorda från en annan. Det första steget fordonet är en mikropartikel tillverkad av biologiskt nedbrytbar poröst kisel (PSI), medan den andra etappen fordon är nanopartiklar laddade med läkemedel eller kontrastmedel 6,7. Nanopartiklarna, som är inbäddade i PSI materialet, gradvis släppt som Si degraderar 8. En fördel med att använda Si-partiklar är att morfologin och ytegenskaperna enkelt kan skräddarsys för att uppnå optimal biofördelning och läkemedelsfrisättning. Nyligen var en framgångsrik användning av MSV plattform för leverans av siRNA liposomer till tumörvävnad som visas i en äggstocks och bröstcancer musmodell 9, 10. I detta arbete har vi tillverkat ett universellt leveranssystem för siRNA baserat på huvudmännen i MSV-plattformen. Effekten av detta leveranssystem har tidigare visats ossing olika siRNA-molekyler 11. Systemet är en polykatjon-functionalized poröst kisel (PCPS) bärare, som består av psi ympad med polyetylenimin (PEI) och arginin (Arg). PEI kan hjälpa till vid bildande av elektrostatiska interaktioner med siRNA, medan Arg och Psi kan tjäna till att reducera toxiciteten av PEI, som tidigare demonstarted 11. Dessutom kan närvaron av PEI hjälpa intracellulär upptag och endosomal fly, medan PSI mikro möjliggör siRNA skydd och fördröjd frisättning. Psi partiklarna successivt försämra under fysiologiska förhållanden, vilket resulterar i bildandet av Arg-PEI / siRNA nanopartiklar (Figur 1), som har en distinkt morfologi och en snäv storleksfördelning 11. Denna PCPS plattform skiljer sig från den konventionella MSV, eftersom andra steget nanopartiklar inte initialt present i bäraren, men bildas med tiden som den första etappen bäraren degraderar 11, 12. Den siRNA lastning effektivitet, cytotoxicitet och gen tysta effektivitet PCPS systemet har utvärderats in vitro. porous silicon microparticles for delivery of sirna therapeutics

Small interfering RNA (siRNA) can be used to suppress gene expression, thereby providing a new avenue for the treatment of various diseases. However, the successful implementation of siRNA therapy requires the use of delivery platforms that can overcome the major challenges of siRNA delivery, such as enzymatic degradation, low intracellular uptake and lysosomal entrapment. Here, a protocol for the preparation and use of a biocompatible and effective siRNA delivery system is presented. This platform consists of polyethylenimine (PEI) and arginine (Arg)-grafted porous silicon microparticles, which can be loaded with siRNA by performing a simple mixing step. The silicon particles are gradually degraded over time, thereby triggering the formation of Arg-PEI/siRNA nanoparticles. This delivery vehicle provides a means for protecting and internalizing siRNA, without causing cytotoxicity. The major steps of polycation functionalization, particle characterization, and siRNA loading are outlined in detail. In addition, the procedures for determining particle uptake, cytotoxicity, and transfection efficacy are also described.
