Dit protocol maakt de snelle verrijking van de zebravis embryonale hartweefsel voor genexpressie analyses. Zebravis (Danio rerio) wordt veel gebruikt in ontwikkelingsbiologie in organogenese in vivo te bestuderen vanwege zijn snelle, transparante en buitenbaarmoederlijke embryonale ontwikkeling, in combinatie met de geringe omvang en de beschikbaarheid van transgene reporter lijnen met weefsel-specifieke expressie van fluorescerende eiwitten. Dit kleine gewervelde is bijzonder geschikt voor hartontwikkeling studeren, omdat oxygenatie van de vroege zebravis embryo niet afhankelijk hartslag en bloedstroom; Deze functies zijn toegelaten de karakterisering van grote aantallen cardiovasculaire mutanten 1,2 en de zebravis is nu algemeen erkend modelorganisme hartziekten 3 bestuderen. Om de genexpressie te bestuderen tijdens de embryonale ontwikkeling, zijn transcripties vaak geanalyseerd door geheel-mount in situ hybridisatie (WISH) 4, RT-qPCR 5, microarrays 6, of next generation sequencing (RNA-Seq) 7. TerwijlWISH kan een ruimtelijk-temporele analyse van genexpressie in het gehele embryo worden transcript niveaus gewoonlijk bepaald door RT-qPCR, microarrays of RNA-Seq benaderingen. Deze werkwijzen vereisen weefsel verrijking voor genexpressie profilering. Omdat het zebravis embryonale hart slechts een klein deel van de gehele embryo, transcriptoom studies gedurende cardiale ontwikkeling vereist een protocol voor dissectie en verrijking van harten. Bovendien fysiologisch relevante gegevens te verkrijgen, is het belangrijk om de cardiale weefsel volledig functioneel tot RNA-extractie behouden. We beschrijven hier een protocol voor het snel isoleren fysiologisch normaal en kloppende harten van honderden zebravis embryo&#39;s van hoge kwaliteit RNA monsters efficiënt te krijgen voor verdere analyses. De onderhavige werkwijze is gebaseerd op het protocol beschreven door Burns en MacRae, 2006 8. Voor verrijking van hartweefsel beide methoden een transgene myocardiale reporter lijnen profiteer van de differentiële hechting eigenschappen van de zebravis embryonale harten versus andere weefsels. Kort pipetteren veel embryo en neer door een smalle pipetpunt, harten gelijktijdig vrijkomen uit embryonale organen en vervolgens gescheiden uit embryonale puin in twee stappen snel filtreren; fluorescent gelabelde harten worden vervolgens handmatig gesorteerd blijft vuil en verzameld voor verdere verwerking.

De zebravis embryonale hart is samengesteld uit slechts een paar honderd cellen, die slechts een klein deel van de gehele embryo. Daarom, om de cardiale transcriptoom van wordt gemaskeerd door het wereldwijde embryonale transcriptoom voorkomen, moet voldoende harten voor verdere analyse te verzamelen. Zoals zebravis hartontwikkeling snel verloopt, hart inzameling en RNA winningsmethoden moet snel zijn om de homogeniteit van de monsters te verzekeren. Hier presenteren we een snelle handleiding dissectie protocol voor het verzamelen van functionele / kloppende harten van de zebravis embryo&#39;s. Dit is een essentiële voorwaarde voor verdere cardiaal-specifieke RNA-extractie cardiale genexpressie te bepalen door transcriptoom analyse, zoals kwantitatieve real-time polymerase-kettingreactie (RT-qPCR). De methode is gebaseerd op differentiële klevende eigenschappen van de zebravis embryonale hart in vergelijking met andere weefsels; Dit maakt een snelle PHYsieke scheiding van cardiale van extracardiale weefsel door een combinatie van vloeibare dwarskracht verstoring stapsgewijze filtratie en handmatige verzameling van transgene fluorescent gelabelde harten.

We beschrijven hier een representatief hart dissectie experiment met behulp van de zebravis Tg (myl7: GFP) twu34 transgene lijn 9, die groen fluorescerend eiwit (GFP) drukt uitsluitend binnen het myocard (afb. 1).
