Методологии, основанные высокопроизводительного секвенирования были широко применяется для многих биологических образцов в последние годы значительно расширяющих наши представления о молекулярной сложности биологических систем 3,4. Тем не менее, подготовка образцов РНК для высокопроизводительного секвенирования часто придает определенные предубеждения, присущие занятого методологии, ограничивающие потенциальную полезность этих мощных методов. Эти метод конкретных предубеждения были хорошо документированы для перевязки основе, малых РНК библиотечных препаратов 1,2,5,6. Эти предубеждения привести к изменению 1000 раз в считывает числа для эквимолярными синтетических микроРНК, делая вывод о микроРНК изобилия из данных секвенирования дико переменная и ошибок.
