WNV is een BL3 pathogeen. Vanwege veiligheidsvoorschriften, worden immunologische tests met WNV besmette monsters vaak beperkt door de beschikbaarheid van apparatuur op BL3 faciliteiten of meer langdurig en vervelend om te presteren. In een recente studie, gebruikten we twee stroomcytometrie gebaseerde methoden om celgemedieerde immuunresponsen bestuderen gedurende WNV infectie 5. In beide assays werden WNV-geïnfecteerde cellen behandeld met 1-2% PFA rechtstreeks of fixatie / permeabilisatie oplossing met 4%...

West-Nijlvirus (WNV), een neurotropisch, plus-voelde flavivirus, is een opkomende bedreiging voor de volksgezondheid. Op dit moment zijn er geen vaccins zijn goedgekeurd voor menselijk gebruik 1. Een verzwakte WNV stam, die een P38G substitutie in het niet-structurele (NS) 4B eiwit, bekend geen letaliteit bij muizen maar hogere aangeboren cytokinen en T cel responsen in muizen dan wildtype WNV NY99 stam 2 induceren. Muizen geïmmuniseerd met de NS4B-P38G mutant werden allemaal beschermd tegen een secundaire uitdaging met dodelijke wild-type WNV. Dit suggereert dat de NS4B-P38G mutant geschikte kenmerken voor een ideaal vaccin kandidaat. De mechanismen waarmee de NS4B-P38G mutant induceert hoge beschermende adaptieve immuniteit is nog niet duidelijk begrepen. Toll-like receptoren (TLR&#39;s), die pathogeen-geassocieerde moleculaire patronen te herkennen, spelen een essentiële rol in de initiatie van aangeboren immuniteit tegen virale infectie. De kern TLR signaalweg maakt gebruik van myeloïde differentiatie primaire reactie gene 88 (MyD88) als de primaire adapter 3,4. In een recente studie werd MyD88 signalering blijkt een belangrijke rol in de ontwikkeling van celgemedieerde immuniteit spelen tijdens WNV NS4B- P38G mutante infectie bij muizen 5. Dendritische cellen (DCs) zijn één van de belangrijkste antigeen presenterende cellen vertonen de unieke capaciteit tijdens virale infectie 6,7 primaire T-cel responsen leiden. CD4 + en CD8 + T-cellen dragen bij tot langdurige beschermende immuniteit en zijn belangrijk voor gastheer overleven na wildtype WNV infectie 8,9. Twee immunologische assays werden bij dit onderzoek naar de functie van deze cellen in de NS4B-P38G mutant geïnfecteerde muizen te beoordelen. Eerst werd een in vitro T-cel priming assay gebruikt ter vergelijking de antigenpresenterende vermogen van DCs van WNV-geïnfecteerde wildtype en MyD88 - / - muizen. Om de gevoeligheid van de assay, naïeve CD4 &lt;vergrotensup&gt; + T-cellen werden geïsoleerd uit OTII transgene muizen, die een Vα2 / Vβ5 TCR specifiek voor de kip ovalbumine (OVA) peptide uitdrukken 323 - 339. DC&#39;s van WNV geïnfecteerde muizen werden gezuiverd, en co-gekweekt met carboxyfluoresceïne succinimidyl Pasen (CFSE ) -gemerkte CD4 + T-cellen in aanwezigheid van OVA-peptide. Na 5 dagen van co-kweek werden de cellen geoogst en met paraformaldehyde (PFA) vastgesteld en door flow cytometrie geanalyseerd. Proliferatieve assays zijn vanouds door incorporatie van 5-broom-2&#39;-deoxyuridine (BrdU) of getritieerd thymine deoxyriboside (3 HTdr) 10 uitgevoerd. Toch zijn deze testen zijn ofwel radioactief en / of die bijzondere apparatuur in bioveiligheidsniveau (BL) 3 faciliteiten waar WNV studies worden uitgevoerd. De flowcytometrische analyse van lymfocyt proliferatie door seriële halvering van de fluorescentie-intensiteit van de vitale kleurstof CFSE geworden vaker gebruikt in immunologische assays de kleurstof stabieleren gelijkmatig opgenomen in cellen, gemakkelijk gedetecteerd door flow cytometrie en is nonradioactive 11. De test heeft ook de mogelijkheid om het aantal celdelingen beoordelen. Een groot voordeel van deze assay in WNV studies is dat fixatie van de geïnfecteerde cellen met 1-2% PFA WNV 12, die monsterverzameling mogelijk maakt met een flowcytometer in BL2 laboratorium kunnen inactiveren. Vervolgens werd een gemodificeerd intracellulaire cytokine kleuring (ICS) procedure gebruikt om de rol van MyD88 signalering in regulatie van WNV specifieke T cel responsen in NS4B-P38G-mutant geïnfecteerde muizen te bestuderen. In deze test werden splenocyten geïsoleerd uit geïnfecteerde muizen behandeld in vitro met WNV specifieke peptiden. Brefeldine A werd toegevoegd aan de cytokinen in de cel behouden. Na 5 uur incubatie werden cellen geoogst, gewassen en gekleurd voor T cel subsets. Cellen werden vervolgens gefixeerd in PFA, gepermeabiliseerd en gekleurd voor interferon (IFN) -γ en geanalyseerd met flow cytometrie.Zoals bij andere flowcytometrie gebaseerde bepaling, wanneer de cellen worden behandeld met fixatie en permeabilisatie buffer met PFA, geïnfecteerde monsters kunnen worden overgebracht naar een BL2 laboratorium voor verdere verwerking en analyse. In verschillende gepubliceerde studies, hebben we gebruik ICS T celeffectorfuncties meten WNV-geïnfecteerde muizen 13,14. Hoewel het goed gevestigd, een belangrijk nadeel van deze test is dat de procedure zeer langdurig en tijdrovend wanneer deze binnen BL3 voorzieningen kunnen zijn. Hier werd een micro-centrifugebuis ICS-methode getoond haalbaarder, gemakkelijker te gaan en minder tijdrovend wanneer deze binnen een BL3 laboratorium zijn.

Een verzwakt West Nile virus (WNV), een niet-structurele (NS) 4B-P38G mutant geïnduceerde hogere aangeboren cytokine en T cel responsen dan het wildtype WNV bij muizen. Onlangs, myeloïde differentiatie factor 88 (MyD88) signalering werd getoond belangrijk voor initiële T-cel priming en geheugen T-cel ontwikkeling te zijn tijdens WNV NS4B-P38G mutant infectie. In deze studie twee stroomcytometrie-gebaseerde methoden - een in vitro T-cel priming assay en een intracellulaire cytokine kleuring (ICS) - werden gebruikt om dendritische cellen (DCs) en T celfuncties beoordelen. In de T-cel priming test werd celproliferatie geanalyseerd door flow cytometrie na co-cultuur van DCs uit beide groepen muizen met carboxyfluoresceïne succinimidylester (CFSE) - gelabelde CD4 + T-cellen van OTII transgene muizen. Deze aanpak gaf een nauwkeurige bepaling van het percentage prolifererende CD4 + T-cellen met aanzienlijk verbeterde algehele gevoeligheid dan de traditieal assays met radioactieve reagentia. Een microcentrifugebuis werd gebruikt in zowel celkweek en cytokine kleuringen van de ICS protocol. Vergeleken met de traditionele weefselkweek-plate-gebaseerd systeem, deze aangepaste procedure was makkelijker om te presteren op de bioveiligheid niveau (BL) 3 faciliteiten. Bovendien werden WNV- geïnfecteerde cellen behandeld met paraformaldehyde in beide assays, die verdere analyse buiten BL3 faciliteiten ingeschakeld. Kortom, kunnen deze in vitro immunologische assays worden gebruikt om celgemedieerde immuunresponsen in WNV infectie efficiënt beoordelen.

In de T-cel priming test CFSE gelabelde CD4 + T-cellen werden gekweekt met gezuiverd DCs uit de NS4B-P38G-mutant geïnfecteerde wildtype en MyD88 - / - muizen in de aanwezigheid of afwezigheid van OVA-peptiden.
