Neste artigo é descrito um método para obter um modelo reprodutível de lesão medular traumática usando um Infinito Horizon Impactor com uma força de 70 Kdyne (grave). Usando um paradigma força maior (80 Kdyne), que pode causar uma lesão mais grave que, infelizmente, está associado à mortalidade camundongos mais elevados. A lesão medular (LM) é a lesão mais comum causada por trauma de alta energia, como veículos automóveis acidentes, quedas, esportes e violência 1. Em grave SCI, a força lesão destrói ou danifica o tecido neural, causando perda súbita da função neurológica. Traumatic SCI ocorre frequentemente em adultos jovens entre 10 e 40 anos de idade. Ela afeta grandemente condição mental e física do paciente e causa enorme impacto econômico para a sociedade 2. A abordagem de tratamento na fase aguda é geralmente limitada a uma alta dose de corticosteróide, a estabilização cirúrgica e descompressão para possivelmente atenuar danos maiores 3-4, mas os papéis desses métodos na recuperação locomotora após SCI ainda são controversos. Além da perda de tecido aguda, lesão traumática e a activação de mecanismos secundários de degeneração causa desmielinização e morte de vários tipos de células 5-6. O grau de recuperação da função pode dedez ser correlacionado com a extensão da matéria branca poupado no local da lesão 7. Os modelos animais de SCI pode ser usado tanto para investigar as respostas biológicas do tecido a lesão e para testar terapias potenciais. Além disso, um modelo animal útil de uma patologia humana, não só tem que reproduzem alguns aspectos dessa condição, mas deve também oferecer vantagens sobre observação clínica directa e experimento. Muitos estudos avaliaram os efeitos de transplante de uma variedade de células-tronco em modelos SCI 11. Temos recentemente isolado adultos neural células-tronco a partir da Sub-Ventricular Zone (SVZ) várias horas após a morte do doador do mouse 12-13. Além disso, neste trabalho demonstram que as células transplantadas migrar para locais de lesão da medula espinhal e diferenciar principalmente em proteína associada microtúbulos (MAP) 2 células positivas. Além disso, a diferenciação é acompanhada pela promoção de uma recuperação estável da função dos membros posteriores.

A lesão medular é uma condição clínica devastadora, caracterizada por um complexo de disfunções neurológicas. Os modelos animais de lesão da medula espinal pode ser utilizado tanto para investigar as respostas biológicas a lesões e para testar terapias potenciais. Contusão ou compressão lesão entregue à medula espinal exposta cirurgicamente são os modelos mais amplamente utilizados de patologia. Aqui demonstramos um método adaptado para lesão da medula espinal, seguido por injecção na veia da cauda de células em ratos CD1. Em suma, nós fornecemos procedimentos para: i) marcação celular with um traçador vital, ii) o atendimento pré-operatório de ratos, iii) execução de uma lesão medular contundente, e iv) a administração intravenosa de pós mortem precursores neurais. Este modelo de contusão pode ser utilizado para avaliar a eficácia e segurança de transplante de células estaminais em uma abordagem de medicina regenerativa.

O número total de células transplantadas é 1 x 10 6 células e foi dividido em três injecções consecutivas na veia da cauda. Nós administrado 3,3 x 10 5 células em 50 ul de solução tampão de fosfato (PBS). A primeira injecção foi realizada no prazo de 30 minutos após a lesão, o segundo 6 horas mais tarde e as últimas 18 horas após a lesão.
