Terminal deoxynucleotidyltransferase (TdT) dUTP nick end labeling (TUNEL) is het proces van het gebruik van de TdT enzym om dUTP tot 3 hechten &#39;einden van de dubbele en enkelstrengs DNA breekt 12,23. De TUNEL methode voor de detectie van apoptose en DNA schade werd voor het eerst 20 jaar geleden door Gavrieli et al. 1,12,24. Het is sindsdien geëvalueerd en geoptimaliseerd in verschillende weefselpreparaten 7,23,27,40. TUNEL is gebruikt om ischemie geïnduceerde celdood van neuronen 6,14,29 en 43,44 cardiomyocyten, excitotoxische neuronale celdood 30,31 bestuderen, en als biomarker in artritisbehandeling 39. Er is ook gebruikt als een prognostische factor en tumorcel marker in verschillende menselijke kankers 2,3,15,32,37,38,42. Alternatieve methoden bestaan ​​voor DNA schade en celdood detectie, maar ze hebben technische uitdagingen en restricties. Southern blotting kan worden gebruikt quantify DNA schade in zijn geheel weefsel lysaten 7,9-11, maar deze methode biedt geen cellulair niveau resolutie en is moeilijk te kwantificeren. De komeettest is een alternatief cel-gebaseerde methode die vereist extraheren bewaarde kernen van cellen 4,20,28,36. Hoewel de comet assay werkt goed op gekweekte geïsoleerde cellen, is het veel moeilijker om intacte kernen bereiden van skeletspierweefsel 8,21. Zoals met Southern blot, ofwel de comet assay niet celtype-specifieke informatie van een hele spierweefsel homogenaat verschaffen. Een ander alternatief voor de TUNEL methode immunohistochemie met antilichamen tegen enkelstrengs DNA 25,33,41 of tegen eiwitten die deelnemen DNA schade respons en celdood routes (bijvoorbeeld p53, H2AX en caspasen) 13,17,22,40. Dergelijke antilichaam gebaseerde methoden vereisen grondige karakterisatie van antilichamen en uitstekende antilichaamspecificiteit een hoge signaal-achtergrond verhouding oplevert. Zelfs wanneer specific antilichamen bestaan, kunnen zij denatureren van het doeleiwit door antigen retrieval procedure 34,35 vereisen. Zoals we hier bespreken, antigen retrieval in spierweefsel leidt tot onaanvaardbaar hoge autofluorescence. In tegenstelling tot de alternatieve methoden, TUNEL behaalt DNA-schade detectie met een hoge signaal en lage achtergrond, uitstekende specificiteit die getest kunnen worden met eenvoudige positieve en negatieve controles, goede weefselpenetratie dat antigen retrieval niet vereist, en cellulair niveau resolutie. Bovendien neemt de TUNEL methode ongeveer 4 uur in beslag, terwijl alternatieve overnacht incubatie vereisen meestal. We bestuderen skeletspier celdood in een muismodel van spinale musculaire atrofie (SMA) 10 die is gegenereerd door Hsieh-Li en collega 16. Apoptotische cellen te kwantificeren in de spier, hebben wij een werkwijze weefselpreparaat, kleuring en kwantificering die robuust werkt in verschillende skele ontwikkeldTal spiergroepen op verschillende ontwikkelingsstoornissen tijdstippen bij muizen. We maken gebruik van een in de handel verkrijgbaar TUNEL-labeling kit en in de handel verkrijgbare software voor beeldanalyse. We hebben ook met succes gebruikt de TUNEL assay in combinatie met immunofluorescente kleuring in het ruggenmerg 10. De hier beschreven methoden zijn nuttig voor onderzoekers die willen weefselpathologie mechanismen van ziekten, mechanismen van veroudering en ontwikkelingsstoornissen (pre- en postnatale) celdood in de skeletspier. De TUNEL-techniek is bijzonder nuttig voor studies van DNA schade en herstel en celdood in modelsystemen waar slechts een subset van cellen beïnvloed en celniveau resolutie noodzakelijk. Deze video beschrijft dissectie, weefsel verwerking, snijden, en fluorescentie gebaseerde TUNEL etikettering van de muis skeletspieren. Terminal deoxynucleotidyl transferase (TdT) deoxyuridine triphosphate (dUTP) nick end labeling (TUNEL) is the method of using the TdT enzyme to covalently attach a tagged form of dUTP to 3&#8217; ends of double- and single-stranded DNA breaks in cells. It is a reliable and useful method to detect DNA damage and cell death in situ. This video describes dissection, tissue processing, sectioning, and fluorescence-based TUNEL labeling of mouse skeletal muscle. It also describes a method of semi-automated TUNEL signal quantitation. Inherent normal tissue features and tissue processing conditions affect the ability of the TdT enzyme to efficiently label DNA. Tissue processing may also add undesirable autofluorescence that will interfere with TUNEL signal detection. Therefore, it is important to empirically determine tissue processing and TUNEL labeling methods that will yield the optimal signal-to-noise ratio for subsequent quantitation. The fluorescence-based assay described here provides a way to exclude autofluorescent signal by digital channel subtraction. The TUNEL assay, used with appropriate tissue processing techniques and controls, is a relatively fast, reproducible, quantitative method for detecting apoptosis in tissue. It can be used to confirm DNA damage and apoptosis as pathological mechanisms, to identify affected cell types, and to assess the efficacy of therapeutic treatments in vivo.
