Vi har beskrevet en strømlinjeformet protokoll som har blitt brukt til å behandle mer enn 1600 fullblodprøver i Detroit Neighborhood Health Study. Disse studiene har imidlertid blitt innhentet fra kliniske settinger der sykdomsassosierte forskjeller kan være forstørret på grunn av den generelt mer alvorlig karakter av sykdommer for hvilke pasienter som søker behandling. På grunn av fremskritt i &quot;omics&quot; nærmer seg, har det siste tiåret sett en eksplosjon av interesse i å få biologiske prøver fra samfunnet og / eller epidemiologiske innstillinger 5-7, for å gi befolkningsbaserte estimater av utbredelsen sykdom og et bredere bilde av miljømessige determinanter av disse mentale og / eller fysiske sykdommer. Protokollen er basert på vår samling og ombygging av dagens teknikker, inkludert lagring av alle &quot;ekstra&quot; fraksjoner i tilfelle fremtidige teknikker tillate ytterligere isolering / analyser. Selv om alternative fremgangsmåter eller sett kan anvendes i stedet for de enkelte fremgangsmåter som er beskrevet her, de som er beskrevet har vist seg å være en pålitelig og effektiv måte for å behandle prøvene i en high-throughput måte. Høykvalitets fraksjoner (PBMC, DNA, serum, og RNA) av friskt blod kan produseres innen 2 timer av samlingen og alle analyse-ready prøvene kan foreligge innen 2 dager (Figur 1). rapid fractionation and isolation of whole blood components in samples obtained from a community-based setting

Innsamling og behandling av fullblodprøver i en ikke-klinisk setting tilbyr en unik mulighet til å evaluere samfunns bolig individer både med og uten preexisting forhold. Rask behandling av disse prøvene er avgjørende for å unngå degradering av viktige cellulære komponenter. Inkludert her er metoder for samtidig perifert blod mononukleære celler (PBMC), DNA, RNA og serum isolasjon fra en enkelt blodprøvetaking utført i hjemmene til samtykkende deltakere over et byområde, med behandlingen initieres innen to timer av samlingen. Vi har brukt disse teknikkene for å behandle over 1600 blodprøver, hvilket ga jevn og høy materialkvalitet, som deretter er blitt anvendt i vellykket DNA-metylering, genotyping, genekspresjon og flowcytometri analyser. Noen av metodene som benyttes er standard; imidlertid, når kombinert på den beskrevne måte, gjør det mulig at de effektiv behandling av prøver fra deltakerne i population- og / eller samfunns-baserte studier som normalt ikke ville bli evaluert i en klinisk setting.
