In vitro haben neuronalen Kulturen als ein grundlegendes Instrument für die Untersuchung von neurologischen Erkrankungen eingesetzt. Primäre tierische (meist Nagetier) neuronalen Kulturen 1, 2 und humanen neuralen Zelllinien aus Gliomen und anderen Tumore abgeleitet sind die am häufigsten verwendete in solchen Studien. Jedoch wurde erkannt, dass es signifikante Unterschiede zwischen Nager- und menschliche Zellen. Viele Erkenntnisse basierend auf Nagetiere können nicht auf den Menschen übertragen werden. Darüber hinaus mit der rasanten Entwicklung bei der Analyse von Massen genomischer Informationen und die relativ einfach Gen Bearbeitung und Sequenzierung ganzer Genome, geht der Trend immer mehr darauf ausgerichtet, die Entdeckung Krankheit anfällig Gene und die Abgrenzung ihrer Funktionen und Rollen in bestimmten Krankheiten, die die wenigen menschlichen neuronalen macht Zelllinien sind nur begrenzt Gebrauch. Theoretisch humanen primären neuronalen Kulturen aus Proben von Patienten Nervensystem abgeleitet werden, sind die beste Wahl, aber sie unmöglich zu erhalten sind; daher alternative meththoden notwendig. Kurze Zeit später wurde ein anderer Ansatz entwickelt, um direkt zu verwandeln neuronalen Zellen aus somatischen Zellen 8, 9. Menschliche Krankheit spezifischen neuronalen Kulturen sind für die Erzeugung von in-vitro-Modellen für neurologische Erkrankungen des Menschen. Der fehlende Zugang zu primären humanen adulten neuralen Kulturen wirft jedoch einzigartige Herausforderungen. Die jüngsten Entwicklungen in induzierte pluripotente Stammzellen (iPS) bietet einen alternativen Ansatz zur neuronalen Kulturen aus Hautfibroblasten durch patientenspezifische iPS ableiten, aber dieser Prozess ist arbeitsintensiv, erfordert spezielles Know-how und große Mengen an Ressourcen und kann mehrere Monate dauern. Dies verhindert, dass die breite Anwendung dieser Technologie auf die Untersuchung von neurologischen Erkrankungen. Um einige dieser Probleme zu überwinden, haben wir eine Methode, um neurale Stammzellen direkt aus menschlichen adulten peripheren Blut abgeleitet entwickelt, unter Umgehung der iPSC Ableitungsprozesses. Hämatopoetischen Vorläuferzellen aus humanen adulten peripheren Blut angereichert wurden in vitro kultiviert und mit Sendai-Virus-Vektoren, die Transkriptionsfaktoren Sox2, Oktober transfiziert3/4, Klf4 und c-Myc. Die Transfektionsergebnisse in morphologischen Veränderungen in den Zellen, die unter Verwendung von humanen neuralen Vorläufer Medium mit basischem Fibroblasten-Wachstumsfaktor (bFGF) und den vaskulären endothelialen Wachstumsfaktor (VEGF) weiterhin ausgewählt sind. Die resultierenden Zellen werden durch die Expression neuronaler Stammzellenmarker wie Nestin und SOX2 gekennzeichnet. Diese Nervenstammzellen könnte weiter zu Neuronen, Astroglia und Oligodendrozyten in bestimmten Differenzierungsmedien unterschieden werden. Mit leicht zugänglichen menschlichen peripheren Blut-Proben, könnte diese Methode verwendet werden, um neurale Stammzellen zur weiteren Differenzierung zu Nervenzellen für In-vitro-Modellierung von neurologischen Störungen abgeleitet werden und können Studien zur Pathogenese und Behandlung dieser Krankheiten zu fördern.
