인간의 질병 특정의 연결을 문화는 인간의 신경 질환에 대한 체외 모델의 생성에 필수적이다. 그러나, 인간 성인 일차 배양 신경에 대한 액세스는 고유의 부족 문제를 제기한다. 유도 만능 줄기 세포의 최근 개발 (IPSC)는 환자의 특정 IPSC을 통해 피부 섬유 아 세포에서 신경 문화를 유도하기 위해 또 다른 방법을 제공하지만,이 과정은 노동 집약적이며, 특별한 전문 지식과 자원의 많은 양을 필요로하고, 몇 달이 걸릴 수 있습니다. 이것은 신경 질환의 연구에이 기술의 다양한 응용 프로그램을 방지 할 수 있습니다. 이러한 문제들을 극복하기 위해, 우리는 IPSC 도출 과정을 거치지 않고, 직접 사람 성인 말초 혈액으로부터 신경 줄기 세포를 유도하는 방법을 개발 하였다. 성인 인간의 말초 혈액에서 농축 조혈 전구 세포가 시험 관내에서 배양하고 10 월을 전사 요인 SOX2 함유 센다이 바이러스 벡터로 형질3/4, Klf4 및 c-Myc와. 상기 염기성 섬유 아세포 성장 인자 (bFGF)와 혈관 내피 성장 인자 (VEGF)를 포함하는 인간 신경 전구 매체를 이용하여 선택되는 세포의 형태 학적 변화의 형질 전환 결과. 생성 된 세포는 네 스틴과 같은 SOX2 같은 신경 줄기 세포 마커에 대한 발현을 특징으로한다. 이러한 신경 줄기 세포는 더 뉴런, 아스트로 글 리아 세포가 지정된 차별화 미디어에서 희소 돌기 아교 세포로 분화 될 수있다. 쉽게 접근 인간 말초 혈액 샘플을 사용하여,이 방법은 신경 질환의 체외 모델링 신경 세포로 더욱 분화 신경 줄기 세포를 유도하는데 이용 될 수 있으며, 이들 질환의 병인 및 치료에 관한 시험을 진행할 수있다.
