Culturas neuronais específicos doenças humanas são essenciais para a geração de modelos in vitro para doenças neurológicas humanos. No entanto, a falta de acesso para culturas neuronais adultas humanas primárias coloca desafios. Desenvolvimentos recentes em células-tronco pluripotentes induzidas (IPSC) fornece uma abordagem alternativa para derivar culturas neurais a partir de fibroblastos da pele através de paciente iPSC específico, mas este processo é trabalhoso, exige conhecimentos especiais e grandes quantidades de recursos, e pode levar vários meses. Para superar algumas destas questões, temos desenvolvido um método para obter células-tronco neurais diretamente do sangue periférico humano adulto, ignorando o processo de derivação da IPSC. As células progenitoras hematopoiéticas enriquecidos a partir do sangue periférico humano adulto foram cultivadas in vitro e transfectadas com vectores de vírus Sendai contendo factores de transcrição Sox2, Out3/4, KLF4, e c-Myc. Os resultados de transfecção em alterações morfológicas nas células, que são ainda seleccionadas usando meio progenitora neuronal humana contendo factor de crescimento de fibroblastos básico (bFGF) e factor de crescimento endotelial vascular (VEGF). As células resultantes são caracterizadas pela expressão de marcadores de células estaminais neuronais, tal como a nestina e SOX2. Essas células-tronco neurais pode ser ainda mais diferenciado para os neurônios, astroglia e oligodendrócitos em media diferenciação especificado. Utilizando amostras de sangue periférico humano facilmente acessíveis, este método pode ser utilizado para derivar células estaminais neurais para uma maior diferenciação para células neuronais in vitro de modelagem de desordens neurológicas e pode avançar estudos relacionados com a patogénese e tratamento dessas doenças.
