De hier beschreven methoden helpen om te illustreren de verspreiding en de complexe cel autonome en niet-autonome cel toxiciteit van prion-achtige eiwitten. We hebben onlangs ontdekt dat een aggregatie-gevoelige cytosole prion domein wordt opgenomen in membraangebonden blaasjes in een autofagie gerelateerde proces. Vele neurodegeneratieve ziekten, waaronder de ziekte van Alzheimer (AD), de ziekte van Parkinson (PD), amyotrofische laterale sclerose (ALS), en overdraagbare spongiforme encefalopathieën (TSE&#39;s), worden geassocieerd met aggregerende eiwitten en daarmee gezamenlijk bekend als eiwit misfolding stoornissen (PMDs ). TSE of prion ziekten vormen een unieke klasse van PMDs doordat zij infectieuze bij mens en dier 1 kan zijn. Op moleculair niveau, prionen repliceren door het aantrekken en het omzetten van monomere α-helix-rijk-gastheer gecodeerd cellulaire PrP (PrP C) in de pathologische β-sheet-rijke Prpsc conformatie 2,3. Zelf propageren eiwit aggregaten zijn ook geïdentificeerd in schimmels, die belangrijke kenmerken van zoogdieren prionen 4,5 delen. Daarnaast zoogdieren prionen kunnen verplaatsen van cel tot cel en infecteren naïeve cellen 6,7. Terwijl PMDs other dan TSE&#39;s zijn niet besmettelijk, ze delen een gemeenschappelijke pathogene principe met prionziekten 8,9. Hoewel de eiwitten verbonden aan elk van de PMDs niet verbonden zijn in structuur of functie, ze vormen aggregaten via een kristallisatieproces-achtige proces genaamd kiemvorming of geënt polymerisatie; bovendien eiwithoudende zaden te laten groeien door het aantrekken van hun oplosbare isovormen 2,10,11. De efficiëntie zichzelf propageren varieert in vivo afhankelijk van de intrinsieke eigenschappen van het eiwit, die samen met andere cellulaire factoren zoals moleculaire chaperones tarieven aggregaat kiemvorming, zaaien, fragmentatie en verspreiding 12-15 uiteindelijk bepalen. Daarom moet er een goede balans tussen deze factoren efficiënte propagatie van eiwit aggregatie toelaat bestaan. Dit zou ook verklaren waarom slechts enkele amyloïdogene aggregaten haven de kenmerken van een prion, en dus niet alle PMDs zijn besmettelijk. Prionen lijken o &#39;top-performers&#39; vertegenwoordigenfa breed spectrum van zelfreplicerende eiwitachtige aggregaten, waardoor ze een krachtig hulpmiddel om PMDs 8,13 studeren maakt. Intrigerend de toxiciteit geassocieerd met ziekte gerelateerde aggregaten vaak niet cel autonoom component 16,17. Dit betekent dat zij invloed naburige cellen die geen expressie het overeenkomstige gen, in tegenstelling tot een strikt cel autonoom effect, wat impliceert dat alleen de cellen die het gen vertonen specifieke fenotype. Deze werd overtuigend aangetoond door weefsel-specifieke expressie of neerhalen van de respectievelijke eiwitten in tal van modellen van neurodegeneratieve ziekten 18-26. Verschillende mechanismen zijn voorgesteld als basis voor deze niet-cel autonome toxiciteit in PMDs, waaronder verminderde toevoer van voedingsstoffen, onbalans in neuronale signalering, glutamaat excitotoxiciteit en neuroinflammation 16,27,28. Bovendien, een prion-achtige beweging van ziekte gekoppelde aggregaten tussen cellen might dragen bij aan dit aspect 29,30. Meer bewijs suggereert dat andere eiwitten insluitsels dan prionen kan overbrengen van cel-cel, wat kan verklaren de karakteristieke verspreiding pathologie waargenomen in vele PMDs 30-36. Echter, het is nog te bepalen of er een duidelijk oorzakelijk verband tussen de intercellulaire beweging van ziekte-eiwitten en het toxische effect op naburige cellen. Daarom is een beter begrip van de cellulaire routes die cel tot cel transmissie en niet cel autonoom toxiciteit grondslag liggen noodzakelijk en essentieel voor de ontwikkeling van nieuwe therapeutica. Maar veel aspecten van prion-achtige verspreiden en cellulaire factoren die cel tot cel transmissie van verkeerd gevouwen eiwitten beïnvloeden metazoa niet goed begrepen, in het bijzonder op organismale niveau. De nematode Caenorhabditis elegans heeft een aantal voordelen die het potentieel heeft ontdek nieuwe facetten van prion-achtige spreading in metazoa 17. Het is transparant, waardoor in vivo volgen van fluorescent gelabeld eiwitten in de levende organisme. Verder veel cellulaire en fysiologische processen die door ziekte zijn geconserveerd tegen wormen menselijke en C. elegans ook geschikt zijn voor een breed scala van genetische manipulatie en moleculaire en biochemische analyses 37-39. Precies 959 somatische cellen vormen de volwassen hermafrodiet met een eenvoudige lichaam plan dat nog steeds heeft een aantal verschillende soorten weefsel, met inbegrip van de spieren, neuronen en darm. Om een nieuwe prion model in C. vestigen elegans, kozen we ervoor om exogeen drukken de goed gekarakteriseerd glutamine / asparagine (Q / N) rijke prion domein NM van de cytosole gist prion eiwit Sup35, aangezien er geen bekende endogene prion-eiwitten in wormen 4,40. Gist prionen zijn van onschatbare waarde in het ophelderen van fundamentele mechanismen van prion replicatie 41-44 geweest. Bovendien, NM is de sparst cytosolische prion-achtige eiwit dat is aangetoond dat de volledige levenscyclus van een prion recapituleren in zoogdiercelkweek 45,46. Ook bij expressie in C. elegans, het opmerkelijk goed vastgesteld aan de verschillende vereisten voor propagatie in metazoan cellen vergeleken met gistcellen vertoonden belangrijkste kenmerken van prion biologie 40. NM aggregatie geassocieerd met een ernstige toxische fenotype, zoals verstoring van mitochondriale integriteit en het uiterlijk van Sup35 prion domein verschillende autophagy gerelateerde blaasjes op cellulair niveau, alsmede embryonale en larvale arrestatie, ontwikkelingsachterstand en een wijdverspreide verstoring van de eiwitvouwing omgeving op het organismale niveau. Opvallend het prion domein vertoont cel zelfstandige en niet cel autonoom toxiciteit, die naburige weefsels waarin het transgen niet tot uitdrukking. Verder wordt de vesiculaire transport van het prion-domein binnen en tussen cellen gecontroleerd real time &lt;em&gt; In vivo 40. Hier beschrijven we hoe u prion-achtige verspreiding in C. onderzoeken elegans. We zullen uitleggen hoe de intra- en intercellulaire transport van blaasjes met daarin het prion domein met behulp van time-lapse fluorescentie microscopie te monitoren. We zullen het gebruik van weefsel-specifieke vouwen sensoren benadrukken en alomtegenwoordig uitgedrukt stress-verslaggevers naar cel autonome en niet-cel autonome effecten op cellulair conditie te bepalen. Ten slotte zullen we de procedure van een onlangs uitgevoerde genoomwijde RNA interferentie (RNAi) scherm te beschrijven om nieuwe modifiers van prion-geïnduceerde toxiciteit identificeren. Tezamen kunnen deze werkwijzen helpen plagen elkaar genetische routes betrokken bij de intercellulaire beweging van eiwitten en hun niet cel autonoom toxiciteit.

Prions are unconventional self-propagating proteinaceous particles, devoid of any coding nucleic acid. These proteinaceous seeds serve as templates for the conversion and replication of their benign cellular isoform. Accumulating evidence suggests that many protein aggregates can act as self-propagating templates and corrupt the folding of cognate proteins. Although aggregates can be functional under certain circumstances, this process often leads to the disruption of the cellular protein homeostasis (proteostasis), eventually leading to devastating diseases such as Alzheimer&#8217;s disease (AD), Parkinson&#8217;s disease (PD), Amyotrophic lateral sclerosis (ALS), or transmissible spongiform encephalopathies (TSEs). The exact mechanisms of prion propagation and cell-to-cell spreading of protein aggregates are still subjects of intense investigation. To further this knowledge, recently a new metazoan model in Caenorhabditis elegans, for expression of the prion domain of the cytosolic yeast prion protein Sup35 has been established. This prion model offers several advantages, as it allows direct monitoring of the fluorescently tagged prion domain in living animals and ease of genetic approaches. Described here are methods to study prion-like behavior of protein aggregates and to identify modifiers of prion-induced toxicity using C. elegans.
