Un épendymaires lignes de la monocouche de cellules du système ventriculaire du cerveau fournissant des fonctions de barrière et de transport bi-directionnelles entre le fluide spinal cérébral (CSF) et de fluide interstitiel (ISF) 3.1. Ces fonctions aident à garder le cerveau des toxiques gratuitement et en équilibre physiologique 2,3. Chez l&#39;homme la perte de portions de ce revêtement sur blessure ou la maladie ne semble pas résulter en remplacement de régénération que l&#39;on trouve dans d&#39;autres revêtements épithéliaux; plutôt une perte de couverture de la cellule épendymaire semble résulter en astrogliose périventriculaire avec un maillage d&#39;astrocytes couvrant des régions dénudées des cellules épendymaires à la surface du ventricule. Les répercussions graves aux mécanismes CSF / d&#39;échange de l&#39;ISF et de dédouanement importants, pourraient être prévues résulter de la perte de cette couche épithéliale 1,2,4-7. Une caractéristique commune de vieillissement humain est agrandie ventricules latéraux (ventriculomégalie) et associé œdème périventriculaire comme observed par l&#39;IRM et la récupération d&#39;inversion fluide atténué IRM (IRM / FLAIR) 8-14. Pour étudier la relation entre ventriculomégalie et l&#39;organisation cellulaire de la muqueuse du ventricule, des séquences d&#39;IRM post mortem humains ont été jumelés à des préparations histologiques de ventricule latéral du tissu péri-ventriculaire. En cas de ventriculomégalie, d&#39;importantes zones de gliose avaient remplacé la couverture cellulaire épendymaire le long de la paroi du ventricule latéral. Lorsque l&#39;expansion du ventricule n&#39;a pas été détecté par l&#39;analyse du volume basé sur l&#39;IRM, la doublure de la cellule épendymaire était intact et gliose n&#39;a pas été détectée le long de la paroi du ventricule 6. Cette approche combinatoire représente la première documentation détaillant les changements globaux dans l&#39;intégrité cellulaire de la muqueuse du ventricule latéral en utilisant des préparations wholemount de parties ou la totalité de la paroi du ventricule latéral et la modélisation 3D des volumes du ventricule 6. Plusieurs maladies (maladie d&#39;Alzheimer, la schizophrénie) et les blessures (traumatismes crâniens)montrer ventriculomégalie comme une caractéristique neuropathologique tôt. Dénudation des zones du revêtement de la cellule épendymaire ainsi serait prévu pour interférer avec la fonction de la cellule épendymaire normale et compromettre l&#39;équilibre homéostatique entre CSF / fluide ISF et l&#39;échange de soluté. Le manque de données d&#39;imagerie multimodaux, et en particulier les séquences de données longitudinales, avec un accès limité à des échantillons de tissus histologiques correspondant fait l&#39;analyse des pathologies du cerveau humain difficile. 3d modeling of the lateral ventricles and histological characterization of periventricular tissue in humans and mouse

The ventricular system carries and circulates cerebral spinal fluid (CSF) and facilitates clearance of solutes and toxins from the brain. The functional units of the ventricles are ciliated epithelial cells termed ependymal cells, which line the ventricles and through ciliary action are capable of generating laminar flow of CSF at the ventricle surface. This monolayer of ependymal cells also provides barrier and filtration functions that promote exchange between brain interstitial fluids (ISF) and circulating CSF. Biochemical changes in the brain are thereby reflected in the composition of the CSF and destruction of the ependyma can disrupt the delicate balance of CSF and ISF exchange. In humans there is a strong correlation between lateral ventricle expansion and aging. Age-associated ventriculomegaly can occur even in the absence of dementia or obstruction of CSF flow. The exact cause and progression of ventriculomegaly is often unknown; however, enlarged ventricles can show regional and, often, extensive loss of ependymal cell coverage with ventricle surface astrogliosis and associated periventricular edema replacing the functional ependymal cell monolayer. Using MRI scans together with postmortem human brain tissue, we describe how to prepare, image and compile 3D renderings of lateral ventricle volumes, calculate lateral ventricle volumes, and characterize periventricular tissue through immunohistochemical analysis of en face lateral ventricle wall tissue preparations. Corresponding analyses of mouse brain tissue are also presented supporting the use of mouse models as a means to evaluate changes to the lateral ventricles and periventricular tissue found in human aging and disease. Together, these protocols allow investigations into the cause and effect of ventriculomegaly and highlight techniques to study ventricular system health and its important barrier and filtration functions within the brain.
