DArgic la signalisation par les neurones dans la région tegmentale ventrale (VTA) et substantia nigra pars compacta de (SNC) du mésencéphale est considéré comme important pour les comportements cognitifs, émotionnels et moteurs récompense motivés. Cependant, la signalisation de DA trop ou trop peu mésencéphale provoque de nombreux symptômes invalidants dans une variété de troubles neurologiques (par exemple la maladie de Parkinson, déficit de l&#39;attention et hyperactivité, schizophrénie et la toxicomanie). Médicaments qui augmentent ou diminuent DA signalisation soulager ces symptômes, mais ils produisent également des effets secondaires attribuables à la signalisation dérégulée et les effets hors-cible. L&#39;efficacité du médicament diminue également au fil du temps en raison des réponses compensatoires du cerveau. Le défi est donc de rétablir la signalisation DA mésencéphale normale, de façon plus ciblée et physiologique, et une approche favorisée est en augmentant ou en diminuant le nombre de neurones dopaminergiques du mésencéphale. La preuve a été accumulé pendant sevsieurs décennies que l&#39;expression des gènes et des protéines impliquées dans le métabolisme et le trafic DA et d&#39;autres catécholamines dans les cellules adultes matures est modifiable (examinés dans une). Dans le mésencéphale, le nombre de tyrosine hydroxylase immunopositif (TH +, l&#39;enzyme limitant la vitesse à Da synthèse) neurones diminue puis augmente suite à l&#39;administration de la neurotoxine 2,3, tandis que le nombre de TH immunonegative (TH) neurones montre la tendance inverse (ce est à dire des augmentations puis diminue 3). Ceci est cohérent avec la perte ensuite gain du «phénotype DA &#39;par certaines cellules. Le nombre de neurones TH TH + et CNS a également été démontré que le changement dans des directions égales mais opposées suivant divers traitements qui modifient l&#39;activité électrique de ces cellules à 4,5. Par exemple, la perfusion de la petite conductance, de potassium activé par le calcium (SK) dans le canal antagoniste apamine mésencéphale pendant 2 semaines diminue le nombre de TH + et augmente (par la même quantité) à la numbre de TH- SNc neurones 4,5. En revanche, la perfusion de l&#39;agoniste des canaux SK-1 EBIO augmente le nombre de TH + et diminue (par le même montant) le nombre de neurones TH SNc 4,5. Des changements similaires ont été observés suite à une variété de traitements ciblant SNc activité neuronale, y compris certains qui visait entrées afférentes 4. Ce règlement apparente du nombre de neurones SNc DArgic par l&#39;activité neuronale et entrée afférente soulève la possibilité que l&#39;environnement ou le comportement peuvent influer sur le nombre de neurones de SNC. En effet, des souris adultes exposés à des environnements différents ont plus ou moins mésencéphale (SNC et VTA) TH + neurones, et au moins certains de ces changements induits par l&#39;environnement sont abolis par le blocage simultané de l&#39;entrée synaptique dans le mésencéphale 6. environmental modulations of the number of midbrain dopamine neurons in adult mice

Des changements durables dans le cerveau ou «plasticité cérébrale» sous-tendent le comportement adaptatif et la réparation du cerveau après les maladies ou des blessures. En outre, les interactions avec notre environnement peuvent induire la plasticité du cerveau. De plus en plus, la recherche tente d&#39;identifier les environnements stimuler la plasticité cérébrale bénéfique pour traiter les troubles cérébraux et comportementaux. Deux manipulations de l&#39;environnement qui sont décrits augmenter ou diminuer le nombre de tyrosine hydroxylase immunopositif (TH +, l&#39;enzyme limitant la vitesse dans la dopamine (DA) synthèse) des neurones dans le mésencéphale de souris adulte. La première comprend appariement souris mâles et femelles de façon continue pendant 1 semaine, ce qui augmente TH mésencéphale + neurones d&#39;environ 12% chez les hommes, mais diminue TH mésencéphale + neurones d&#39;environ 12% chez les femmes. La seconde comprend les souris de logement en continu pendant deux semaines dans des «environnements enrichis» (EE) contenant roues de roulement, des jouets, des cordes, des matériaux de nidification, etc., qui increases TH mésencéphale + neurones d&#39;environ 14% chez les hommes. En outre, un protocole est décrite pour infuser simultanément des médicaments directement dans le mésencéphale au cours de ces manipulations de l&#39;environnement pour aider à identifier les mécanismes sous-jacents de l&#39;environnement induite par la plasticité du cerveau. Par exemple, EE-induction de plus TH mésencéphale + neurones est abolie par le blocage simultané de l&#39;entrée synaptique sur les neurones du mésencéphale. Ensemble, ces données indiquent que l&#39;information sur l&#39;environnement est relayé via l&#39;entrée synaptique des neurones du mésencéphale pour allumer ou éteindre l&#39;expression de gènes «DA».
