Traditionele benaderingen voor toxicologische tests op uitgebreide dierstudies waardoor testen kostbaar en tijdrovend. Bovendien kan door de interspecies verschillen preklinisch veiligheid bij dieren niet altijd geldig toxische effecten van potentiële geneesmiddelen relevant voor de mens voorspellen. Hoewel niet-menselijke primaten zijn meest voorspelbare, nog steeds sterk ethische en socioeconomical eisen snel verhogen door de moderne samenlevingen voor het ontwikkelen van gevoelige en robuuste in vitro-test ...

Het vermogen van humane embryonale stamcellen (hESC) te differentiëren tot verschillende typen cellen opende een nieuw tijdperk van in vitro toxiciteitsproeven 1, ziektemodel en regeneratieve geneeskunde 2. De stamcellen zijn begiftigd met de capaciteit om te repliceren, om hun pluripotente toestand in stand te houden en te differentiëren tot gespecialiseerde cellen 3,4. De eigenschappen van hESC (capaciteit om te differentiëren tot alle belangrijke celtypes) zijn ook gevonden in andere menselijke pluripotente stamcellen, zoals de mens geïnduceerde pluripotente stamcellen (iPSC) of cellen die door nucleaire transfer 5. Zo hebben vele verschillende cellijnen gedifferentieerd tot neuronen 6, 7 niercellen, neurale cellen 8, cardiomyocyten 12/09 of hepatocyten achtige cellen 13,14. Bovendien kan hESC spontaan differentiëren in cellen van alle drie de kiem lagen 15-18 in embryoid lichamen (EBS) 19,20. Early embryonale ontwikkeling wordt gereguleerd door differentiële expressie van verschillende genen betrokken bij de verschillende kiem lagen die op mRNA-niveau is vastgelegd door transcriptomics behulp van microarray technologie 15. Deze inspanningen resulteerden in de oprichting van het orgaan specifiek toxicologisch modellen gebaseerd op hESC / iPSC en transcriptomics analyse (zie voor een overzicht 21,22). Deze modellen hebben voordelen ten opzichte van het traditionele gebruik van proefdieren voor toxicologische studies, zoals preklinische studies met proefdieren zijn niet altijd voorspellend voor de menselijke veiligheid. De drug geïnduceerde toxiciteit aangetroffen bij patiënten zijn vaak gerelateerd aan metabole of signalering processen die verschillen tussen mensen en proefdieren. Het verschil soort is de betrouwbare vroege opsporing van ontwikkelingsstoornissen voorkomen bij de mens, en bijvoorbeeld drugs zoals thalidomide 23,24 en 25,26 diethylstilbestrol werden uit de markt genomen vanwege teratogeniciteit. ThaliDomide heeft niet aangetoond geen ontwikkelingsstoornissen bij ratten of muizen. Milieuchemicaliën zoals methylkwik 27 resulteerde in prenatale ontwikkelingstoxiciteit opzichte van het zenuwstelsel in verscheidene species, maar menselijke manifestaties zijn moeilijk te modelleren bij dieren zijn. Om het probleem van soortspecificiteit kwesties aan te pakken, wetenschappers werken onder verschillende projecten op basis van stamcellen zoals reprotect, ESNATS, detective etc. zijn bezig met de ontwikkeling van verschillende modellen voor embryonale toxiciteit, neurotoxiciteit, cardiotoxiciteit, hepatotoxiciteit en nefrotoxiciteit met menselijke toxische stoffen verdacht invloed op de mens. Onder het Europese consortium project &#39;embryonale stamcel-gebaseerde roman van alternatieve testmethoden (ESNATS) hebben vijf testsystemen vastgesteld. Een testsysteem de zogenaamde UKK (U niversitäts k linikum K OLN) testsysteem gedeeltelijk vangt vroege humane embryonale ontwikkeling. In dit system menselijke embryonale H9 cellen worden gedifferentieerd tot drie kiembladen (ectoderm, endoderm en mesoderm) 15 en kiemlaag specifieke handtekeningen zijn gevangen genomen door transcriptomics profiel met de Affymetrix microarray platform. Verschillende ontwikkelings toxische stoffen zoals thalidomide 28, valproïnezuur, methylkwik 16,17 of cytosinearabinoside 15 werden getest in dit systeem en de toxische specifiek gen handtekeningen zijn verkregen. In een tweede test-systeem, de zogenaamde de UKN1 (U niversiteit van K onsta n z) testsysteem 1, wordt H9 cellen gedifferentieerd om neuroectodermale voorlopercellen (NEP) voor 6 dagen. Dit blijkt uit een hoge expressie van neurale gen merkers zoals PAX6 en OTX2. Tijdens de differentiatie gedurende 6 dagen, NEP-cellen blootgesteld aan ontwikkelingsstoornissen neuro-toxische stoffen zoals VPA, methylkwik. Giftige stof-specifieke-de gereguleerde transcriptomics profielen obtai geweestevenals ned met behulp van de Affymetrix microarray platform 16,29. De nieuwe visie voor toxicologie van de 21e eeuw wordt ervan uitgegaan dat testsystemen niet alleen fenotypische beschrijvingen opleveren zoals histopathologie in vivo, of transcriptoom veranderingen aan het eind van de lange termijn giftige stof incubaties. Het eerder suggereert dat assays bieden mechanistische informatie 3, en dat deze informatie kan worden toegewezen aan de zogenaamde negatieve uitkomst paden (AOP), dat een wetenschappelijke rationale voor gevaarlijke effecten 30. Om dergelijke informatie te verstrekken, de toegepaste testsystemen moeten worden zeer kwaliteit gecontroleerd 31, zoals bijvoorbeeld gedocumenteerd door robuuste standaard operatie procedures. Bovendien moeten tijdsafhankelijke veranderingen in kaart te brengen met een hoge resolutie. Dit vereist testsystemen met gesynchroniseerde veranderingen 32. De hier beschreven UKN1 en UKK testsystemen zijn geoptimaliseerd voor deze eisen.

human pluripotent stem cell based developmental toxicity assays for chemical safety screening and systems biology data generation

Efficiënte protocollen menselijke pluripotente stamcellen aan verschillende weefsels in combinatie onderscheiden met -omics technologieën opende nieuwe perspectieven voor in vitro testen van de toxiciteit van potentiële geneesmiddelen. Een solide wetenschappelijke basis voor dergelijke assays verschaffen, is het belangrijk om kwantitatieve informatie te verkrijgen over het tijdsverloop van de ontwikkeling en de onderliggende regulerende mechanismen systeembiologie benaderingen. Twee tests zijn daarom hier afgestemd op deze eisen. In het UKK testsysteem worden menselijke embryonale stamcellen (hESC) (of andere pluripotente cellen) links spontaan differentiëren 14 dagen in embryoid organen, opwekking van cellen van alle weefsels mogelijk. Dit systeem recapituleert belangrijkste stappen van vroege menselijke embryonale ontwikkeling, en kan voorspellen mensspecifieke vroege embryonale / teratogeniciteit, indien cellen worden blootgesteld aan chemicaliën tijdens differentiatie. Het UKN1 testsysteem is gebaseerd op hESC differentiëren naar een population van neuro-ectodermale progenitor (NEP) cellen gedurende 6 dagen. Dit systeem recapituleert vroege neurale ontwikkeling en voorspelt vroege ontwikkelingsneurotoxiciteit en epigenetische veranderingen veroorzaakt door chemicaliën. Beide systemen, in combinatie met transcriptoom microarray studies, zijn geschikt voor het identificeren van biomarkers toxiciteit. Bovendien kunnen zij worden gebruikt in combinatie om data systeembiologie analyse te genereren. Deze testsystemen hebben voordelen ten opzichte van de traditionele toxicologische studies waarbij grote hoeveelheden dieren. De test systemen kan bijdragen tot een vermindering van de kosten voor de ontwikkeling van geneesmiddelen en chemische veiligheidsevaluatie. Hun combinatie belicht vooral op verbindingen die neurodevelopment specifiek kunnen beïnvloeden.

Methylkwik exposure in UKK testsysteem De cytotoxiciteit bepaling werd uitgevoerd met H9 EBS een IC 10-waarde (vermindering van de levensvatbaarheid van 10%) voor de cytotoxiciteit van methylkwik (Fig 1) te verkrijgen. We voerden ook een microarray gebaseerde (Affymetrix platform) biomerker studie. De H9 EBS blootgesteld aan methylkwik (0,25 en 1 uM) gedurende 14 dagen.
