Der Zweck dieses Berichts ist es, ein sicheres und reproduzierbares Verfahren zur Implantat TEVs von Interesse in der Schaf- Halsschlagader ist. Das Hauptaugenmerk bei der Entwicklung TEVs war es, einen Ersatz für autologes Transplantat Ersatz bieten, wenn autologe Gefäße sind nicht verfügbar und die Gebersicht Morbidität zu begrenzen. Beispielsweise hat die Zahl der koronaren Arterien-Bypass-Operationen pro Jahr in den USA 350.000 überschritten, und die ideale Quelle geeigneter Transplantate bleibt der linken Arteria mammaria interna, linke vordere absteigende Koronararterie und die Vena saphena 1. Da viele Menschen, die unter Gefässkrankheiten leiden, nicht geeignet Arterien und Venen für autologes Transplantat ausgetauscht werden müssen, hat die Entwicklung von TEVs somit ein intensives Forschungsfeld werden seit Jahrzehnten 1-6. Während die Konstruktion und Optimierung von neuen TEVs haben viele Fortschritte durchgemacht, die Berichterstattung über die Operationstechniken eingesetzt, um die TEVs sich nicht ein Thema von so intensiven Diskussion gewesen zu implantieren. Vielmehr werden Protokolle über die Implantation TEVs in Tiermodellen weitgehend linksbis zu Ermittler forschen. Die folgende Manuskript zeigt, wie TEVs durch die Verwendung einer End-zu-End-Anastomose Ansatz zu implantieren. Dieses Verfahren wurde unter Verwendung eines spezifischen anastomotischen Nahtmuster, Stabilisierungsnahttechnik optimiert Längsspannung und der Zugabe von in-vivo-Überwachung Instrumentierung optimiert. Dieses Verfahren wird bei einigen der vielen Variationen, die zuvor verwendet wurden, gegenübergestellt. Darüber hinaus wird beschrieben, wie Sie diese Parameter wie arterieller Blutdruck, TEV Durchmesser / Compliance erwerben und Durchflussrate durch das TEV nach der Operation bis Explantation. Die Entwicklung von Tissue Engineered Vessels (TEVs) wird durch die Fähigkeit vorgeschoben, um routinemäßig und effektiv Implantat TEVs (4-5 mm Durchmesser) in einen großen Tiermodell. Ein Schritt-Schritt-Protokoll für interposition Platzierung des TEV und Echtzeit-Digital Beurteilung des TEV und nativen Karotiden wird hier beschrieben. Durch die Implantation von Flusssonden, Katheter und Ultraschallkristallen (fähig In-vivo-Überwachung ermöglicht wird Aufzeichnungs Änderungen implantierter TEVs und nativen Karotiden) zu der Zeit der Operation dynamisch Durchmesser. Nach der Implantation können Forscher arterielle Blutströmungsmuster, invasive Blutdruck und Arteriendurchmesser ergibt Parameter wie Pulswellengeschwindigkeit, Augmentationsindex, Pulsdruck und Einhaltung zu berechnen. Datenerfassung unter Verwendung eines einzelnen Computerprogramms zur Analyse der gesamten Dauer des Experiments durchgeführt. Solche unschätzbaren Daten gibt Einblick in TEV Matrixumbau, dessen resemblance, native / Schein-Kontrollen und Gesamt TEV Leistung in vivo.

Mehr als 30 Schafe, die in diesem Bericht für die Implantation von TEVs beschrieben (im Druck) Operationstechnik 9 unterzogen. Eine Tabelle mit einer Zusammenfassung der jüngsten Schafe Operationen nach Protokolloptimierung sind in Tabelle 2 dargestellt.
