Here we describe in detail a method for inducing PION in a rat model. The most critical part of the protocol is the exposure and irradiation of the optic nerve &#8211; to expose the nerve as long as possible while avoiding damage caused by the sharp fine tip forceps or from stretching. In rats, the ophthalmic artery enters the optic nerve &#8804;1 mm from the optic nerve head. Bij patiënten boven de 50 jaar oud, ischemische optische neuropathie (ION) is de meest voorkomende vorm van acute opticusneuropathie 2. De aandoening kan presenteren als een van de twee subtypes volgens de bron van specifieke aangetaste bloedvoorziening en klinische presentatie: anterior (AION) of zitvlak (PION) 3. Hoewel de pathogenese en verloop van AION zijn uitvoerig bestudeerd 4-7 is PION bleef slecht begrepen vanwege de lage prevalentie variabele presentatie slecht gedefinieerde diagnostische criteria en het ontbreken van een diermodel. Verder zijn er geen behandelingen bewezen effectief te voorkomen of reverse visie verlies van AION of PION. Daarom is een reproduceerbaar en betrouwbaar diermodel van PION is van grote waarde voor het ziekteproces in vivo bestuderen en testen van nieuwe therapeutische strategieën voor neuroprotectie en axon regeneratie. Fotochemisch geïnduceerde ischemische schade aan de microvasculatuur gevolg vasogENIC oedeem en trombose effectief creëert regionale weefselischemie 8-12. Na injectie in het vasculaire circulatie, de fotogevoelige erythrosine B produceert reactieve singlet moleculaire zuurstof bij aktivering met laserbestraling van doelschepen. De singlet zuurstof direct peroxidizes het vasculaire endotheel, het stimuleren van bloedplaatjes hechting / aggregatie en leidt tot de vorming van trombus occlusief. Ischemische schade wordt verspreid naar aangrenzende gebieden en verder verergerd door microvasculaire compressie vanwege vasogeen oedeem. De algemene doelstelling van dit protocol is te fotochemisch ischemie in de retrobulbaire oogzenuw bewegen om de schade veroorzaakt door PION weerspiegelen. Voor zover wij weten is dit het eerste model van ischemisch letsel in het achterste oogzenuw 1. Aangezien dit model produceert ischemie terwijl het vermijden van fysieke trauma&#39;s, de fysiologische processen van de achterste ischemische optische neuropathie zijn beter nagebootst en studeerde. Ook dit model heeft een nieuw platform voor de screening van kandidaat-therapeutica voor optische zenuwaandoeningen en andere CNS ischemische ziekte. Hier, een gedetailleerd protocol voor dijader catheterisatie, optische zenuw blootstelling, intraveneuze injectie van Erythrosin B en laser bestraling in een rat PION model worden beschreven.

Achterste ischemische optische neuropathie (PION) is een sight verwoestende ziekte in de klinische praktijk. Echter, de pathogenese en natuurlijke historie gebleven slecht begrepen. Onlangs ontwikkelden we een betrouwbaar, reproduceerbaar diermodel van PION en geteste het behandelingseffect van bepaalde neurotrofe factoren in het model 1. Het doel van deze video is om onze fotochemisch geïnduceerde model van posterior ischemische optische neuropathie te tonen, en om de effecten ervan met retrograde etikettering van retinale ganglioncellen evalueren. Na chirurgische blootstelling van de achterste oogzenuw, een fotogevoelige kleurstof, erythrosine B, intraveneus geïnjecteerd en een laserbundel wordt gefocusseerd op de optische zenuw oppervlak. Fotochemische interactie van erythrosine B en de laser tijdens bestraling schade het vasculaire endotheel, wordt gevraagd microvasculaire occlusie gemedieerd door trombocytentrombose oedemateus en compressie. De resulterende ischemische letsel levert een geleidelijke, maar pronounced retinale ganglioncellen afsterving, wegens het verlies van axonale ingang - een afstandsbediening-geïnduceerde verwonding en klinisch relevant resultaat. Aldus Dit model verschaft een nieuw platform om de pathofysiologische loop van PION bestuderen, en kunnen verder worden geoptimaliseerd voor het testen van therapeutische toepassingen voor optische neuropathieën en andere CNS ischemische ziekten.

De resulterende ischemie geïnduceerd door deze techniek levert een geleidelijke maar uitgesproken dood van retinale ganglioncellen na ischemisch letsel axon.
