Perinatale Asphyxie ist eine akute und oft unvorhergesehene Zustand mit hypoxisch-ischämischen Enzephalopathie (HIE) zugeordnet ist. Das übergeordnete Ziel dieses Protokolls ist es, eine Ferkelüberlebensmodell der perinatalen hypoxischischämischer Enzephalopathie zu demonstrieren. Der Grad der induzierten HIE ist wesentlich. Ein nützliches Tiermodell der HIE muss auch für die Erprobung neuer dia ermöglichengnostischen Verfahren und Behandlungsmöglichkeiten. Um dies zu ermöglichen, sollten die Modelle moderate HIE, wo es eine Behandlung Potenzial als schwere Hirnschädigung mit wenig oder keiner Behandlung Potenzial wäre weniger relevant bei der Bewertung neuer Behandlungen zu induzieren. Toleranz gegenüber Hypoxie variiert beträchtlich zwischen den Versuchstieren. Frühere Studien haben gezeigt, dass eine konsequentere Hirnschädigung erreicht werden kann und dass mehr Tiere überleben 17,19 durch Individualisierung des induzierter Hypoxie nach Hirnantwort jedes Ferkel durch Amplituden integriert Elektroenzephalographie (aEEG) bewertet und nicht mit einem Satz FiO2-Wert in der gesamten hypoxische Ereignis. Die Dauer aEEG Unterdrückung korreliert mit dem Grad der Gehirnverletzung, mit wenigen histopathologischen Veränderungen bei &lt;20 min aEEG Unterdrückung und schwere Krämpfe Erhöhung bei&gt; 45 min aEEG Unterdrückung. Eine kürzlich durchgeführte Überprüfung der neuroprotektive Behandlungen für HIE identifiziert die Notwendigkeit für das Überleben Modelle ermöglichen Verhaltens Ergebnis measnahmen im Tiermodell 20. Es gibt zahlreiche Vorteile des vorgestellten HIE Ferkelmodell. Es basiert auf einer Art, bei der Ergebnisse mit hoher Wahrscheinlichkeit auf die menschliche Physiologie zu übersetzen beruht. Globale Hypoxie-Ischämie-Modellen Multiorganversagen und Titration von Hypoxie-Ischämie durch aEEG induziert eine konsequente Grad der Hirnschädigung mit dem Überleben klinisch relevante Ergebnisse, dass Biomarker, MRI und Verhalten kann bei entsprechenden Zeitpunkten ausgewertet werden. Piglet Modelle perinatale Asphyxie und HIE haben nicht nur wesentlich zur aktuellen Einblick in HIE Pathophysiologie, sondern auch erfolgreich voran klinischen Studien, was letztlich zu neuen Therapien bei Menschen. Piglet Modellstudien spielte eine Schlüsselrolle bei der Schaffung Hypothermie als Behandlung für HIE 21 und werden in Wiederbelebung von Neugeborenen Forschungs 22 verwendet. Verschiedene Gruppen haben Ferkel-Modelle verwendet werden, wenn die Durchführung von Untersuchungen innerhalb Asphyxie und HIE eind Studien umfassen Hypothermie 23, alpha-Melanozyten-stimulierendes Hormon 24, Herzstillstand 25, Tyrosin-Hydroxylase-Aktivität 26, wiederholt hypoxischen Exposition 27, NMDA-Rezeptor-Aktivität 14 und Nahinfrarotspektroskopie 28. Die Ferkel HIE Modell in diesem Bericht ist technisch anspruchsvoll, mit zu arbeiten, als kleinere Anpassungen im Laufe des Verfahrens kann in entweder zu leicht oder zu schwere Hirnschädigung 29,2 führen. Wir haben festgestellt, dass die vorhandene Literatur fehlte ausreichend detaillierte zuvor veröffentlichten Modelle reproduzieren. So haben wir hier zeigen, jeden Schritt der technischen Verfahren für die Einrichtung eines Ferkel 72 Stunden Überlebensmodell in diesem Bericht erforderlich, die Forscher, um dieses erweiterte Modell für die Untersuchung von HIE etablieren.

Asphyxie unter der Geburt, die hypoxisch-ischämischen Enzephalopathie (HIE) verursacht, entfallen 0,66 Mio. Todesfälle weltweit jedes Jahr rund ein Viertel der weltweit 2,9 Millionen Todesfälle bei Neugeborenen. Tiermodelle von HIE haben zum Verständnis der Pathophysiologie in HIE beigetragen hat und den dynamischen Prozess, der in Hirnverletzung auftreten aufgrund perinataler Asphyxie hervorgehoben. So haben Untersuchungen an Tieren ein Zeitfenster für die Post-Beleidigung Behandlungsstrategien vorgeschlagen. Hypothermie hat sich als Behandlung für HIE in pdiglet Modellen getestet und anschließend in klinischen Studien nachgewiesen wirksam. Variationen des Modells sind in der Studie von Zusatz neuroprotektive Verfahren und Ferkel Studien von Xenon und Melatonin angewendet worden sind, um die klinische Phase I und II führten Studien 1,2. Die Ferkel HIE Modell wird weiter für neonatale resuscitation- und hämodynamische Studien sowie in Untersuchungen von zerebrale Hypoxie auf zellulärer Ebene verwendet. Es ist jedoch eine technisch anspruchsvolleModell und Variationen in dem Protokoll kann entweder zu leicht oder zu schweren Hirnschäden führen. In diesem Artikel zeigen wir die technischen Verfahren für die Schaffung eines stabilen Ferkel Modell der neonatalen HIE notwendig. Zuerst wird die neugeborenen Ferkel (&lt;24 h alt, Durchschnittsgewicht 1500 g) anästhesiert, intubiert und in ein ähnlicher Aufbau wie in einer Intensivstation gefunden überwacht. Globale Hypoxie-Ischämie durch Absenken des inspiratorischen Sauerstofffraktion, globale Hypoxie, Ischämie durch Hypotonie und eine flache Spur Amplitude integriert EEG (aEEG), der zerebrale Hypoxie erreichen induziert. Überleben durch Anpassung Sauerstoffversorgung nach dem aEEG Antwort und der Blutdruck vorangetrieben. Hirnverletzung durch Histopathologie und Magnetresonanztomographie nach 72 h quantifiziert.
