Epitoopaanhangsels zoals HA, Myc, vlag, GST en worden veel gebruikt voor biochemische analyse van eiwitten. Ubiquitination en sumoylation laat de vervoeging van ubiquitine en Kleine Ubiquitine-achtige modifier (SUMO; Smt3 in S. cerevisiae 1) tot een doelwit eiwit, respectievelijk. PTMs van kinetochore eiwitten invloed op hun cellulair niveau en eiwit-eiwit interacties tijdens de verschillende fasen van de celcyclus trouw chromosoom segregatie te garanderen. Zo zijn cellulaire niveaus van Cse4 / CENP-A en buitenste kinetochoor eiwit Dsn1 geregeld door ubiquitine gemedieerde proteolyse te zorgen genoomstabiliteit 2-5. Destabilisatie van onjuiste kinetochoor-microtubule bijlagen vereist de IPL1 / Aurora B kinase, die Dam1 en Ndc80 complexen die rechtstreeks interageren met microtubules 6-8 fosforyleert. Ondanks de identificatie van meer dan zeventig kinetochore eiwitten, zijn er weinig studies die de wijzigingen van deze eiwitten met PTM&#39;s, bijvoorbeeld ubiquitine en SUMO onderzoeken. Een belangrijke beperking is de mogelijkheid om de PTM bewarens tijdens de zuivering en het gebrek aan aangepaste antilichamen voor detectie van PTMs zoals sumoylation, fosforylering, methylering, en anderen. Karakterisering van sumoylated kinetochoor eiwitten Ndc10, Cep3, Bir1 en Ndc80 gebruikt een aangepaste antilichaam 9. Daarnaast is Ndc10 geïmpliceerd als een substraat voor ubiquitinering 10. Human Hec1 (Ndc80 in S. cerevisiae) wordt ook substraat voor ubiquitinering, gereguleerd door APC / C-hCdh1 E3 ligase 11. Daarom Ndc10 en Ndc80 zijn goede kandidaten voor de optimalisatie van het protocol bij zowel sumoylation en ubiquitinering te detecteren in S. cerevisiae. Om de identificatie van sumoylation faciliteren, we geconstrueerd stammen die HF-Smt3 en Myc-gelabeld Ndc10 of Ndc80 uiten. Het gebruik van epitooplabels (HF: His6-vlag) minimaliseert de achtergrond door cross-reactiviteit die vaak wordt waargenomen bij polyklonaal serum opgewekt tegen een kandidaat eiwit. We bedachten een protocol affiniteitzuiveren HF-Smt3 conjugaten en vervolgens gebruikt commerciële anti-Flag en anti-Myc antilichamen om de aanwezigheid van sumolyated Ndc10 en Ndc80 in het gezuiverde preparaat Smt3 detecteren. Voor ubiquitination, we bedacht een aangepaste immunoprecipitatie protocol dat ubiquitinatie van de Myc-gelabelde kinetochoor eiwitten bewaart en uitgevoerd western blot analyse met commerciële anti-Ub antilichaam aan ubiquitinering van Ndc10 en Ndc80 detecteren.

Post-translationele modificaties (PTM), zoals fosforylering, methylering, acetylering, ubiquitinatie en sumoylation, regelen de cellulaire functie van verschillende proteïnen. PTMs van kinetochore eiwitten die associëren met centromere DNA bemiddelen trouw chromosoom segregatie naar genoom stabiliteit te handhaven. Biochemische benaderingen zoals massaspectrometrie en Western blot-analyse worden meestal gebruikt voor de identificatie van PTMs. Hier wordt een eiwitzuivering werkwijze beschreven die de detectie van zowel sumoylation en ubiquitinering van het kinetochore eiwitten, Ndc10 en Ndc80, in Saccharomyces cerevisiae toelaat. Een stam die uitdrukt polyhistidine-Flag-gelabeld Smt3 (HF-Smt3) en Myc-gelabeld Ndc10 of Ndc80 werd gebouwd en gebruikt voor onze studies. Voor detectie van sumoylation, we bedacht een protocol om affiniteit te zuiveren His-tag sumoylated eiwitten met behulp van nikkel kralen en gebruikt western blot analyse met anti-Myc antilichaam aan sumoylated Ndc10 een detecterennd Ndc80. Voor detectie van ubiquitinatie, ontwierpen we een protocol voor immunoprecipitatie van Myc-tagged eiwitten en gebruikt western blot-analyse met anti-Ub-antilichaam aan te tonen dat Ndc10 en Ndc80 worden ubiquitinated. Onze resultaten tonen dat epitoop-gelabeld eiwit van belang in de His-vlag hebben Smt3 stam vergemakkelijkt de detectie van meerdere PTMs. Toekomstige studies moeten toelaten benutting van deze techniek eiwit interacties die afhankelijk zijn van een specifieke PTM identificeren en te karakteriseren.
