Ubiquitinierung und sumoylation ermöglicht die Konjugation von Ubiquitin and Small Ubiquitin-like Modifier (SUMO; Smt3 in S. cerevisiae 1) an ein Zielprotein auf. PTMs der Kinetochor-Proteinen beeinflussen ihre zellulären Ebenen und Protein-Protein-Interaktionen in verschiedenen Zellzyklusphasen treu Chromosomensegregation zu gewährleisten. Um die Identifizierung von Sumoylierung zu erleichtern, konstruierten wir Stämme, die HF-Smt3 und Myc-tagged Ndc10 oder Ndc80 auszudrücken. Die Verwendung von Epitop-Tags (HF: His6-Flag) minimiert den Hintergrund aufgrund von Kreuzreaktivität, die häufig in polyklonales Serum gegen einen Kandidaten-Protein erzeugt wurden beobachtet. Wir entwickelten ein Protokoll zur Affinitätsreinigen HF-Smt3 Konjugate und dann verwendet kommerziellen anti-Flag und Anti-Myc-Antikörper, die Anwesenheit von sumolyated Ndc10 und Ndc80 im gereinigten Präparat Smt3 detektieren. Post-translationale Modifikationen (PTMs), wie Phosphorylierung, Methylierung, Acetylierung, Ubiquitinierung und sumoylation, regulieren die zelluläre Funktion von vielen Proteinen. PTMs der Kinetochor-Proteinen, die mit Centromer-DNA assoziieren vermitteln treu Chromosomensegregation auf Genomstabilität zu gewährleisten.
