Utvecklingen av mikroarrayer, nästa generations sekvensering, och QRT-PCR erbjöd möjligheten att jämföra genuttrycksprofilerna av neuronala populationer i olika biologiska sammanhang 1,2. Dessa känsliga analyser kräver exakt identifiering och isolering av celltyper av intresse samtidigt hålla RNA integritet 2-4. Fluorescensaktiverad cellsortering (FACS) teknik har använts i stor omfattning för att isolera specifika celltyper baserade på markörer och / eller morfologi på cellytan. Här visar vi ett detaljerat förfarande för att få hög kvalitet RNA från en särskild undergrupp av celler i en komplex vävnadsstruktur genom att kombinera snabb immunomärkning med LCM. Vi visar hur du utför viktiga kritiska åtgärder för att få maximal RNA återhämtning och undvika RNA nedbrytning eftersom det kan SIGligt slag genuttryck profilering. För demonstration av detta protokoll, var dopaminerga neuroner från en-dag-gamla mus mitthjärnan riktade. Dopaminerga neuroner kan immunolabeled användning av en antikropp riktad mot tyrosinhydroxylas (TH), det hastighetsbegränsande enzymet för dopaminsyntes. Efter TH immunfärgning, enskilda eller grupper av dopaminerga nervceller kan sedan isoleras med hjälp av LCM. Microdissected Cellerna uppsamlas i lysbuffert, och RNA extraheras med användning av ett RNA-isoleringskit. Kvalitet och kvantitet av extraherat RNA mäts med hjälp av en bioanalyzer 5. Ytterligare analys av genuttryck använder: RNA-sekvensering, microarray, eller QRT-PCR kan därefter utföras 2,4,6. Laser capture microdissection (LCM) möjliggör isolering av specifika celler från tunna sektioner vävnad med hög rumslig upplösning. Effektiv LCM kräver exakt identifiering av celler subpopulationer från en heterogen vävnad. Identifiering av celler av intresse för LCM är vanligtvis baserad på morfologiska kriterier eller fluorescerande protein reportrar. Kombinationen av LCM och snabb immunomärkning erbjuder ett alternativt och effektivt sätt att visualisera specifika celltyper och att isolera dem från omgivande vävnad.
