Auch direkt Kultivierung isolierten primären Darmzellen auf unterschiedlichen Steifigkeit Polyacrylamidgelen noch nicht gemeldet. In den letzten Jahren wurde es immer deutlicher, dass eine mechanische Mikroumgebung, zusätzlich zu bio-chemische Faktoren spielt eine wichtige Rolle bei der Regulierung der Zellfunktionen. Zellen können Erkennen und Reagieren auf das Substrat Steifigkeit auf dem sie bis 3-7 eingehalten (wie in 2D Kultivierung) oder umgeben (wie in 3D-Kultur). Dadurch können die Zellen ihre Differenzierung 3, Morphologie 4, Migration / Motilität 5, biophysikalischen Eigenschaften 6. Wachstums 7 und andere Prozesse zu modulieren. Krebszellen in den 2D und 3D-Matrix Steifigkeit mit einem koordinierten hierarchischen mechano-chemische Kombination von Adhäsionsrezeptoren und assoziierte Signaltransduktion Membranproteine, die Zytoskelett Architektur und molekulare Motoren 1, 2 reagieren auch z. B. Brustepithelzellen (MEC) bilden normale acinar Parenchym, wenn auf 150 Pa Substraten kultiviert, die ähnlich der Steifigkeitvon gesundem Brustgewebe. Interessanterweise weisen sie die Merkmale eines Entwicklungs Tumor, sowohl strukturelle als auch Transkriptions, wenn auf steifer Untergründen (&gt; 5000 Pa), die die Steifigkeit eines Tumorstroma 8 imitieren kultiviert. Zusätzlich zeigt ein weiteres Experiment, dass die Brusttumorgenese durch Kollagenvernetzung und ECM Versteifung 9 verbunden. Kürzlich durchgeführte Experimente zeigen, dass menschliche Kolonkarzinom (HCT-8-Zellen) anzuzeigen Metastasierung Phänotyp (MLP), wenn sie auf Substrate mit 2D physiologisch relevanten Steifigkeit (20-47 kPa) kultivierten, jedoch nicht auf sehr steif (3,6 GPa) Substrate 10- 12 .Diese Zellen erste Form tumorartige Zellcluster dissoziieren dann voneinander, ausgehend von der Peripherie. Da diese epithelialen abgerundete morphologischen (E bis R Übergang) Änderung auftritt, sie vermehren, zu reduzieren Zell-Zell- und Zell-ECM-Adhäsion und werden Migrations. HCT-8-Zellen, die auf sehr harten Polystyrol-Substrate weisen nicht diese kultiviertbösartige Züge. So wurde die Hypothese aufgestellt, dass HCT-8-Zellen werden metastasierendem aufgrund ihrer Exposition gegenüber geeigneter Mikroumgebung. Es ist erwähnenswert, dass diese Versuche werden mit immortalisierten Krebszelllinien oder primäre Zellen murine abgeleiteten, nicht mit primären menschlichen Krebszellen durchgeführt. Eine neue Studie schlägt vor, dass Augmented zellulären Zugspannung kann als Potential biophysikalischen Signatur für metastatischen Zellen 13 .Die Untersuchung umfaßt das Messen Zugkraft für verschiedene menschliche Krebszelllinien auf Polyacrylamidgelen verwendet werden. Es wird festgestellt, dass metastatische Krebszellen deutlich höhere Zugspannung im Vergleich zu nicht-metastatischen Zellen in allen Fällen 13 auszuüben. Allerdings sind diese Ergebnisse direkt im Widerspruch zu den früher veröffentlichten Ergebnisse auf murine abgeleitet Brustkrebs-Zelllinien 14. Auch eine aktuelle Studie zeigt deutliche Unterschiede zwischen immortalisierte und primäre menschliche Zellen in ihrer Zytoskelett Umgestaltung proProtein-Profiling und das Überleben der Zellen Protein-Expression 15. Daher ist es wichtig, viele der biophysikalischen Untersuchungen, wie Traktion für primäre menschliche Krebszellen zu überdenken. Dies wird auf die Frage eingegangen, ob die Primärzellen rekapitulieren immortalisierten Krebszelllinien Traktion Trend. Das hier beschriebene Protokoll wird für die Isolierung von primären menschlichen Darmzellen (beide gesund und Krebs), und für die Kultivierung sie auf weichen Substraten (Polyacrylamid-Hydrogele) sowie auf Petrischalen optimiert. Das Protokoll basiert auf der Verdauung und daraus enzymatische Dissoziation des chirurgischen Gewebeprobe in Einzelzellsuspension 16 basierend. Unseres Wissens ist dies die erste Demonstration der Kultivierung isolierten primären Dickdarmtumorzellen und normalen Zellen direkt auf weiche Hydrogel-Substrate mit integrierten Leuchtstoffmikroperlen für Traktion Zytometrie. Transparentes Gel Substrate erlauben auch Immunfärbung. Dieser Test zeigte Unterschiede in der F-Actin-Organisation undfokalen Adhäsionen in primären menschlichen Darmzellen als Substrat Steifigkeitsänderungen. Diese Zellkultur-Plattform eröffnet die Möglichkeit zu erforschen biophysikalischen Eigenschaften von primären humanen Zellen, wie Zelle Steifigkeit und Traktion als Parameter für die Krebs Prognostik.

Krebszellen reagieren auf mechanische Steifigkeit in komplexer Weise die Matrix unter Verwendung eines koordinierten hierarchischen mechanochemische System Adhäsionsrezeptoren zusammensetzt und assoziierten Signaltransduktion Membranproteine, die Zytoskelett Architektur und molekulare Motoren 1, 2. Tastsinns von verschiedenen Krebszellen in vitro vorwiegend mit immortalisierten Zelllinien oder primäre Zellen murine abgeleitet, mit primären humanen Krebszellen untersucht. Daher ist wenig über die Mechanosensitivität primärer humaner Kolonkarzinomzellen in vitro bekannt. Hier wird ein optimiertes Protokoll entwickelt, das die Isolierung von primären menschlichen Darmzellen von gesunden und kanzerösen chirurgischen menschlichen Gewebeproben beschrieben. Isoliert Doppelpunkt Zellen werden dann auf weichen (2 kPa Steifigkeit) und steif (10 kPa Steifigkeit) Polyacrylamid-Hydrogele und Polystyrol (~ 3,6 GPa Steifigkeit) Substrate durch eine extrazelluläre Matrix funktionalisiert (Fibronektin erfolgreich kultiviertin diesem Fall). Fluoreszierende Mikrokügelchen werden in weiche Gele in der Nähe der Zellkulturoberfläche eingebettet ist, und Traktion Test wird durchgeführt, um zelluläre Kontraktionsspannungen mit dem kostenlosen Open-Access-Software zu bewerten. Außerdem Immunfluoreszenzmikroskopie auf verschiedenen Substraten Steifigkeit liefert nützliche Informationen über primäre Zellmorphologie, Zellskelett Organisation und Vinculin enthält fokalen Adhäsionen als Funktion der Substratsteifigkeit.

Das oben beschriebene Protokoll ist erfolgreich für mehrere Gewebeproben (n = 12) von vier verschiedenen Patienten unter Richtlinien der Ethikkommission eingesetzt. 1A veranschaulicht eine repräsentative kolorektalen Tumor direkt nach der Operation aus dem die Gewebeschnitte für Zellkulturen gewonnen werden. Isolation of Primary Human Colon Tumor Cells from Surgical Tissues and Culturing Them Directly on Soft Elastic Substrates for Traction Cytometry

A protocol is described to extract primary human cells from surgical colon tumor and normal tissues. The isolated cells are then cultured on soft elastic substrates (polyacrylamide hydrogels) functionalized by an extracellular matrix protein, and embedded with fluorescent microbeads. Traction cytometry is performed to assess cellular contractile stresses.
